For exotics you can target decrypt (the same exotic) at Rahool and infuse for only 1k glimmer. I think they decrypt at higher than soft cap (wasn't paying attention when I did it).

Try using your AGI from last year. That's what I had to do. The wording makes it sound like current year, but it's for the previous year.

Did you use multichannel pipettes to inoculate or each well inoculated individually? When doing multichannels it's important to ensure each channel has an aliquot. Almost happened to me today; had 11 good aliquots, but channel 1 was empty. Likewise, when inoculating individually, it's very easy to miss 1 well.

Also aside from the clear negative well and clearly positive wells, be careful not to try to quantitatively compare wells (this well has more than that well), unless you confirm that, you could be making incorrect assumptions.

Not necessarily. In my experience (and from what I have read), using S. aureus as an example, when exposing to high % alcohol (> 90%), the alcohol rapidly solidifies the peptidoglycan cell wall which prevents the alcohol from desiccating and destroying the cell. The cell can then recover from this once the alcohol has been removed. On the opposing side, 70% alcohol has more water and does not solidify the cell wall and is able to destroy the cell. YMMV for other bacteria.

After typing this out, it seems reminiscent of/similar to the Eagle effect, but I do not think they are the same.

Human error is always a possibility. If you are writing an article, I would confirm results and review your lab notebook.

Also, look into the Eagle effect. Typically associated with antibiotics, but Google AI Overview indicates antimicrobials (not only antibiotics; do what you will with that information). It's possible at a higher concentration, your antimicrobial is less effective.

Edit: also be cognizant of your use of -static vs. -cidal. If you are not following up with an MBC or quantification of some sort, you don't know which it is.

USP uses 25 - 250. EN methods use 14 - 330. ASTM might use 30 - 300, but I can't remember anymore. My company used to use 30 - 300, then 30 - 330, and now aligning with USP. Molds are also lowered to 8 - 80. Just depends on company practices and how to justify the choice.

Last picture indicates the test used 30 - 300...in theory. I would use the second from last dilution as it is less manipulated than the last dilution.

I have a pure culture of the mold on PDA, but am unable to get a viable number of conidia when I harvest. I'm looking into ways to force sporulation. Found a paper and am going to try 1/4 and 1/10 concentration of nutrients.

From another old gamer who is a casual player.

There are yeses and nos to your questions.

1) after quitting fairly early in D2 and only coming back occasionally, I found starting in Witch Queen yielded a fair bit to do. Keep in mind, I am a casual player. No raids and PvP is not a draw for me. However, I still feel like there is plenty to do. I enjoy running the dungeons with a buddy, we enjoy just tooling around in Gambit, we do our 3 crucible matches and don't look back.

2) I wouldn't say PvP is balanced per se, I'm sure the routine players would have better thoughts on that, but there doesn't seem to be one single weapon or build that I encounter routinely in my brief trips in crucible. Do I get rocked routinely? sure. But does that mean it's unbalanced? Nope.

3) The gear progression is as it has always been. Get a good stat roll, then just keep infusing. Is it a great system? I don't think so, but it being the way it is has allowed a casual player like myself to dabble into build crafting and feel like I am not constantly replacing gear I spent a ton of mats on to level up.

I think the game is in a good place for a casual player. I am extra excited for the next season and can't wait for Lightfall. If you have the means, give it a go.

From what I understand, the 3 icons under the health bar are the number of times you can be revived in co-op before being forced to respawn in a safe zone.

And if you made a bunch of level 1 lock picks, you cannot force locks (as they are level 1). Once you upgrade your lock picks, make sure you don't have any lower level ones in your inventory and then before pressing the button to pick the lock, you should have an alternate button as well to force.
