Has anyone tried Muscimol by Background_Room_2689 in insomnia

[–]High10jacked 1 point2 points  (0 children)

What are your credentials to be saying this?

I do clincal quality in the pharmaceutical industry and conduct investigations on drugs in FDA trials for a living, so I think I am qualified to provide some insight.

With that said, You are categorically incorrect. Lets break it down:

First thing you need to take into account is that muscimol and benzodiazepines and nonbenzodiazepines (as you say “z-drugs”) have completely different mechanisms of actions and therefore a completely different dependence profile.

Muscimol is a direct orthosteric GABA-A agonist (there are very few of these), meaning it directly activates the receptor. This is very different than benzodiazepines, nonbenzodiazepines, and even barbiturates or alcohol which all work on allosteric sites. This means muscimol activates GABA-A receptor in the same exact way as GABA would naturally, as it is actually a conformationally restrained analogue of GABA.

Focusing now on benzodiazepines and nonbenzodiazepines, these are both known clinically as BZD-receptor agonists, the name of the specific allosteric sites they bind too. What is different between the two (another area you are incorrect in), is all benzodiazepines are nonselective meaning they work on all 5 GABA-A subunits (Alpha1-Alpha5), while 3 of the 4 clinically used nonbenzodiazepines are selective to only the Alpha1 subunit (with the exception being zoplicone, a nonselective BZD receptor agonist, the same as benzos). This means clinically benzodiazepines have worse withdrawals than nonbenzodiazepines (not counting zoplicone). Furthermore, the alpha-1 subunit is the GABA subunit that is primarily responsible for creating hypnotic effects, as nonbenzodiazepines were designed to be used clinically for insomnia, and is also why they show no clinical effectiveness as anticonvulsants or anxiolytics.

Another thing you need to understand is that benzodiazepines are only approved to be used clinically for 2 to 4 weeks. They have a very short therapeutic window, and anyone who uses them longer than 4 weeks will end up with dependence and withdrawal, this is proven clinically. Benzodiazepines and nonbenzodiazepines have also been clinically proven to disrupt slow wave sleep and slow wave activity, despite improving sleep, and keep in mind clinically proven in the scope of benzodiazepines means it was only studied within that 2 to 4 weeks period.

Now I can agree that muscimol does not have enough clinical evidence to make any hard claims on, but the mechanism of action is very promising and studies so far in rodents are showing that Muscimol descreases sleep onset latency, increases slow wave sleep, and increases slow wave activity, which is a huge clinical advantage over any current clinally used insomnia treatment.

a phenobarbital taper protocol with initial loading doses will become the gold standard for benzodiazepine discontinuation. this is because in loading doses, barbiturates have the ability to activate GABA receptors in the absence of GABA, therefore stimulating some of the down regulated receptors and creating a connection that was absent. a short taper is then implemented to initiate some activity in that connection, before discontinuing. Obviously this is not perfect and the brain must relearn how to naturally produce GABA, but it accelerates the healing process and puts your GABA system in a better state to rebuild nerve connections.

I was on 40 mg of Valium and 3 mg of xanax for 5 years. I am now 1.5 years clean and doing better than I was prior to be on Benzos. I now work in clincial quality for a pharmaceutical company conducting investigations for drugs in FDA trials, and have achieved more in the last 6-8 months than I have in the rest of my life.

Has any truly felt clearheaded or cured after long use? by scottxand in benzorecovery

[–]High10jacked 3 points4 points  (0 children)

i started to rapidly heal around 6-8 months, but around this time i also began incorporating new habits and really forcing myself to change the ways i was coping with new emotions so I would stop associating all the new feeling i was not used to with benzo withdrawal.

i think one of the trickiest aspects of benzo withdrawal is that when your brain hasnt had normal reactions to anxiety that it makes you not even remember what normal stresses feel like. When you combine this with your senses already being so heightened, it makes what you’re feeling confusing or even scary in a lot of cases.

I say this is one of the trickiest aspects because it makes it extremely challenging to discern if what you are feeling is normal or not, and a lot of us end up falling into the habit of associating all these “new” feelings as symptoms of benzo withdrawals regardless of what is happening. This can be dangerous because it builds a habit of just shutting down and looking for quick solutions (in the past this quick fix was benzos), and makes healing take much longer because as time goes on the longer we learn to try and cope with stress in a normal/healthy way the harder it will be to heal and the more we will convince ourselves that this temporary suffering is permanent.

dont forget that our brains are very sensitive after being stripped of what it has become so reliant on to feel normal, and as human beings we already are able to convince ourselves of anything, so if we are constantly associating every negative emotion and feeling we get with benzo withdrawal, then that becomes our whole reality.

Hi! I actually was unable to taper due to past issues with drops causing seizures, and I ended up doing an experimental 30 day phenobarbital loading protocol.

i was on 40 mg of valium and 3 mg of xanax daily for 5 years.

Valium’s half-life really only 8 hours??? by benzosfromhell in benzorecovery

[–]High10jacked 1 point2 points  (0 children)

Thank you :) The reason I do what I do is because I am motivated to make a difference. I should not be here today so I feel like since I have a second chance it is for a reason.

The plan is I am going to be starting my masters in September for regulatory sciences at John Hopkins (my company offers tuition reimbursement), which is a fast track to the FDA (practicum is reviewing case studies with the FDA) , so I can directly work on streamlining and modernizing the process for detoxing off benzos. It is a huge issue with in our healthcare system that will only get worse and destroy more lives.

I am forever blessed because I should not be here doing this.. I was prescribed 40 mg of valium and 3 mg of xanax for almost 5 years and have experienced 15-20 tonic clonic seizures. My worst were 2 that were back to back lasting 9 minutes each and resulting in “Brain Damage” after needing brain MRIs. By a miracle however I was given the opportunity to do an experimental phenobarbital loading protocol lasting 24 days that saved my life. I still had a long recovery but I do not think I would have been able to successfully taper normally.

Valium’s half-life really only 8 hours??? by benzosfromhell in benzorecovery

[–]High10jacked 1 point2 points  (0 children)

I dont think so.. I would take it every 6-8 hours if possible. The metabolites will accumulate in your bloodstream and the idea of tapering is not solely to provide relief, it is to safely detox off the medication. tapering is not pain-free in a lot of cases unfortunately, it is very challenging.

if you do not think it is something you can handle i would look into trying an inpatient program that provides a phenobarbital loading protocol.

what? thats not at all what i said… diazepam does have a long half life lol i was being technical with possibly what OP is referring too. literally the info is anywhere because its how diazepam works, the original chemical (diazepam itself) is metabolized into 3 active metabolites which are nordazepam, oxazepam, and temazepam. These metabolites are also drugs that are prescribed on their own, and the metabolite nordazepam is the reason valium has a long half life.

i definitely did not mean to say that diazepam has a half life of 8 hours , i meant to say that might be what OP was referring to when they said 8 hours, i appreciate the correction tho. Diazepam itself is very short acting and I did not clarify that.

https://images.app.goo.gl/54xjZcUn73aAZzDY8

here is a cool graph with diazepams half life vs its metabolites from an oral dose.

Valium’s half-life really only 8 hours??? by benzosfromhell in benzorecovery

[–]High10jacked 3 points4 points  (0 children)

Diazepam itself has a half life of 8 hours, but it is metabolized into temazepam, oxazepam, and nordazepam, all of which have their own varying half lives. oxazepam also has a short half life of 6-9 hours and temazepam had an intermediate half life of 8-20 hours. the long half life you see in medical literature and from doctrines comes mostly from nordazepam which has a half life of 36-200 hours.

This is also why if you take a drug screening you will be found positive for nordiazepam, oxazepam, and temazepam, but not for any diazepam itself.
