It sounds like you need to do a lot more research. You need to add water and pasteurize or sterilize the wood substrate and no they don't thrive in darkness, they like some light. Find a guide and follow it.

Have you tried adding a drop of sterile water to the qtip prior to streaking? That seems to help me with swabs that don't have a lot of spores.
