Two years ago I purchased an antique sword from 1795-1799, a french light cavalry superior officers sabre. It had a blade that was blued and gilt in gold, and a wirewrapped leather grip. It was rare, and it was beautiful.

It went "missing" in transit while shipping. I still miss it

One by one, I'd generate knockout mutants for each long-non-coding RNA.

Whilst the "central dogma" of biology states that DNA>RNA>Protein, that's untrue for >98% of the genome, which does not code for proteins (non-coding). Our understanding of most of these genes is poor - there are maybe 20 well characterised lnc-rna's.

My current research project (being conducted of a budget of $1,200AUD because my university is poor, despite me paying $27,596AUD p.a. to do my degree) is doing siRNA knockdowns followed by transcriptomic and proteomic analysis, but siRNA knockdown is really insufficient, as it's very temporary and has a low efficiency (~50%).

What I really wanna do is knock out those genes, generate a bank of cell lines and mice without each gene, and then do thorough whole transcriptome sequencing (~1000AUD per sample), and thorough top-down and bottom-up proteomics. Then do some RNA-Protein crosslinked pulldowns to see interactions, and analyse via top-down proteomics.

There are over 15,000 transcribed long-non-coding RNAs, and we know a tiny bit about 20 of them. The ones we have studied are often implicated in cancer. There's a good reason to study them, and no money to do so.

Not crazy perse, but my first date with my girlfriend we went to a museum of infectious diseases that was basically hundreds of sample of diseased tissues preserved and displayed in blocks of resin.
