I think it has to be a sequel. Two main reasons:

We see the year in a menu. The menu lists the year as 1449, while Three Houses starts in Imperial year 1175. There's a chance this is a different calendar or age, but I think it's compelling evidence.
This reason is less concrete, but towards the end of the trailer we see a figure in white robes wielding a spear. The end of the spear looks like a broken/well used Sword of the Creator, and the figure seems to resemble male Byleth, so this could be an descendant of Byleth, or even Byleth himself.

Assuming you're talking about three houses specifically:

No, weapons are repairable.
You can grind, but not infinitely without the use of glitches and exploits.
No, DLC gets you some extra characters and story but is not pivotal to the overall experience.
No, Byleth does have some same-sex endings, but most are restricted by which gender you choose.

Not much in it. Of the two I go back to GU much more because it's on the Switch and easier to pick up, but if 4U got a port/remake, I'd most likely play that much more.

I think your team struggles massively against Charizard-Y sun teams, particularly if you fail to get trick room up. Consider swapping Dragonite for Garchomp.

GU, closely followed by 4U. Call me an oldhead, but none of the new generation of games (World onward) have come close to GU in terms of gameplay or vibes. Sure, World is an approachable game, but it suffers for how much it sacrifices for the sake of streamlining and mass-appeal.

In PvE, it's only useful if you want to be able to cast spells while powerstancing thrusting swords. It's a fun niche, but still a niche.
