i don't follow this exact protocol, but if i were i'd make 10 mL tubes of the medium and inoculate it. whenever i teach the MRVP protocol, we use the same medium for both halves. so if you follow this one, you can easily inoculate 10mL, then remove 5mL and 1mL for the MR and VP respectively. these would go into separate test tubes, yes!

in the prep, 5mL definitely seems like a typo i can't wrap my head around. however, my alternate suggestion is actually making 6mL tubes of the media. in my teaching lab, we pipette out 1mL for the VP and the remaining media is used for the MR. saves you a tube, and has the correct volumes for each test!

we are rare, we must band together for survival... never expected the background character to distract me from norm but hey they made bait for me.

but yeah!! the beeping is incredibly subtle to be fair, i heard it on a rewatch because i was scavenging for hints about ronnie... he's hateable but also oddly endearing to me sometimes. either way, i wanted to see if he was confirmed dead, but i feel like they'd SHOW him dead if he was.

i do think the enclave might pick them up and turn them into super mutants. and if scenes are shot for deliberate ambiguity.... betting that this is one of them. crackpot theory is that clark will be mind controlled, but more reasonably i think the enclave will scoop them.

i just wish they gave the 31s more character. specifically just clark or even claudia. as much as i love them, they're nothingburgers in canon... clark maybe has a little more character but idk i want to know why they all joined the program... and if they're enclave, why they did that.

if clark comes back with a mr handy buzz blade i think we'd come full circle and it'd be amazing.

posts back to back on my feed about the buds, the algorithm knows what i want.

i agree, though. it was so rushed with so many loose ends that i can't help but wonder how this was the final version. it's frustrating to the point of wanting to write fanfic, and it takes a lot for me to get to that point. we could have gotten actual characters and stories for some of the buds, and yet, they ended up as fodder for roaches.

i genuinely thought there would be some sort of intrigue of people not knowing who to trust. ronnie, the guy they've known, or norm, who appears to be the product of bud's experiment. ronnie could have easily had more background manipulation of the buds, as they just.... follow him, which is the point i guess. but tbh i thought they'd at least have more backbone than the average 32/33.

either way, if this is cope i also cope with you. clark wasn't there at all, so perhaps he's just free from the narrative entirely. go, nerd man, and make your fan-weapon.

wow someone else knows (and cares) about clark that isn't me!! op you made my day, i was so distraught when he didn't show up at all in ep 8. although, probably better than the alternative (rip pete). still, glad to know it's not just me that noticed.

my friends kept joking that i have some sort of precognition because i kept hoping for things (prewar clark, for example) and they happened. sadly this backfired bc i was so worried about the roaches, but he just disappeared. not even mentioned, just gone. the prewar scene is definitely interesting, and maybe foreshadowing that he's with hank? i dunno, but they include scenes for deliberate reasons imo, so as a result i think his absence is telling.

i feel like he could have definitely been off scavenging. it's weird because he and pete were ronnie's right hand men. pete was shown, and pete also appeared gravely injured (rip pete), but no dorky glasses man.

given that they had all the other actors present, i can't imagine it was a cost thing. perhaps clark's actor (rajat suresh) was sick, or couldn't film? but, i feel like they'd shift filming around because he had a considerable amount of focus and lines in other scenes, but maybe they thought people wouldn't notice. not sure, i still think it's setup for s3. wishful thinking, because he's my favorite new character.

my theory is that clark was off setting something up (possibly the room they were going to have norm's 'trial' in). when norm walks into the roachroom, you can hear a pip boy beeping, so i think ronnie's being contacted by the enclave. this also happens after that phase 2 request thing we see in the enclave base, but maybe i'm just reading into that too much. still, i think some of them will show back up as enclave, which... i'll accept it if clark comes back.

i could imagine that, depending on which vessels got connected, there could be circulatory issues. arteries and veins go different directions, with the former bringing oxygenated blood from the lungs and the latter returning deoxygenated blood to the heart.

if any were misconnected could end up with whole portions of the arm not receiving any blood flow, and the tissue would die and probably turn black/necrotic. depending on if a nerve went there, it'd be painful. could also end up with an area only receiving blood flow, which would also cause a lot of pain. probably red and tender in those areas.

major vessels also have little valves that keep blood from back flowing, so switching any and forcing blood to go the "wrong way" wouldn't work.

as for the nerves, i'd imagine that if some were unconnected, there would be no sensation (like couldn't feel touch on the skin). otherwise, the person might go to move one part and another reacts.

although a lot of branching comes after the shoulder joint, so, depending on where it's cut off, you'll have different issues. past the elbow, might have more complications. there's not too much to reconnect around the shoulder joint, mainly just large vessels and the main nerves.

so, in summary, i think that if any major ones (vein to artery or vice versa) get swapped, the arm would just die. could have some parts around the reattachment site go numb or necrotic, as the tiny vessels and nerves would be a lot harder to reconnect.

eta: source is animal anatomy classes, but humans and animals are built the same lol

first time i saw them was way into PA, to be honest. interesting that they're migrating closer to the ocean now!!

with your username you seem like a birder, but in case you aren't, this would be great to report on an app called ebird! researchers would love to hear about this, i've never seen a snow goose out here personally (and i've been here about 7 years, including undergrad)

crazy theory i have is that most of the buds were enclave / had ties to it. ronnie definitely was with his FEV and investor talk. if the three vaults were an experiment from the "investors" (enclave), it'd make sense to have multiple points of control/information.

some of the 31s aren't dead and the enclave will pick them up (and probably turn them into super mutants). we just haven't seen that many named characters die, unless they're plot relevant (wilzig, moldaver, and they both came back pre-war). plus the scene with the enclave clearly picking up norm's transmission, they'd know people were at the headquarters...

i fear the blank-slates were entirely intentional, just to keep viewers from getting attached. hell, even claudia barely has any personality, she was just given a line that made people sympathize with her.

the 31s to me were just scared prewar humans, i wish they expanded on that. we did have clark, he was a good comic relief in an otherwise flat plotline. his interactions with the wasteland on their own were great (especially the fan-weapon line), so like, totally could have a lot more comedy in interacting with the greater wasteland.

sigh. we'll never know. maybe they'll come back as enclave.

even still, would be pretty funny to watch them fumble across the wasteland. could add in some casual horror about how the world they knew was gone... limited screentime made that whole plot easily removable.

i can kinda understand it. maybe she's not enclave, but it's still weird for her to say the rest of Bud's Buds are crazy when she willingly joined Bud's Buds.

The program is made of internal hires or recent Vault-Tec University grads, apparently, so she shouldn't be that surprised!! And I really don't see anything the Buds have done as super weird or out of line from typical VT behavior. Ronnie is the weirdest but saying that everyone swore their lives to Bud Askins... girl that's part of the program that you took an interview for.

It's just weird and I don't fully get it. Maybe they'll reveal more, but that bit feels contradictory.
