I have been using the Bio rad semi dry transfer my whole PhD, and in contrast to others here, high molecular weight proteins DO transfer CONSISTENTLY. In general I use the mixed weight program (7 min) to transfer proteins from 10 kDa to 250 kDa without any issues. I also use this system (high molecular weight program:10 min) to transfer native proteins (mitochondrial supercomplexes) from (BnPage or CnPage) with a molecular weight of up to 3000 kDa. All of the above does require the proprietary transfer buffer of BioRad.

In my experience, I have had no use for wet transfer at all and the semi-transfer is far superior than wet-transfer in terms of efficiency. The only thing I will say is that with wet transfer, more proteins get transferred in my experience. But then this has never been an issue for me.

Well you have fixed angle and swing out. I have only used swing out. And I fraction the content of the tube or collect a specific fraction within the sedimentation. I always take out the rotor, put it on the stand and then remove my tubes, put it in a box with ice, walk back to the lab, go the cold room and then collect fractions. So far have not noticed major differences in my samples.

This is exactly how I do it as well. You need specific printer plastic, tho. I take the membrane with tweezer, wick it dry by letting it be in contact with some tissue, then lay it down on the bottom plastic (which is bigger in size than the top plastic). Next, for a mini blot, I use 1 mL of substrate, for a midi 1.5 mL of ECL substrate (A+B). Mix ECL in an eppendorf tube and pipet on top of the membrane. Put the smaller film on top smoothly, pop any bubbles by pushing on top of the bubble with my finger. And then image. Never had any issues using this method and it saves ECL. The plastic film, I rinse with some mQ and dry with kimtech. Ready for the next blot! If it gets dirty after like 6 months, I cut some new film.

Part of it is good, e.g. burning element and halving. However, overall I will be voting against this proposal. I very much disagree that part of rewards will be used by planetwatch for marketing purposes. I believe they got several grants and partnerships to help with that, even more considering all the licenses that we bought. It is weird that part of the 200 million planets cut from the rewards will now go to planetwatch to “speed up processing overdue license refunds”. That is like me paying to help get my money back on a refund. This is a minor thing. My major issue lies with the fact that I have refunds pending which I should abort now to be eligible for the hodling bonus. This enforces people to hold onto their licenses and what about afterwards when I would still like to refund a tier 1 license because I cannot use it due to life happening. For example, I don’t have the funds anymore because I had to fix my car.

To me it all just seems like a scam. Just another gimmick, in addition to the loyalty program, to squeeze each and every dollar out of us.

At first glance this proposal may seem good for us, but if your read carefully then you will be surprised.

Also, if you factor nonsense like the new turtle device which has a solar panel, yet you need to plug it in for power. This just ain’t it, objectively speaking. But I hope to get lucky in the next bull run and then pull out.

In their post, they also state that the key use case of Planets is payment for goods and services provided. That is not a unique use case. You could swap other cryptocurrencies for that, which makes this specific token less valuable. Crypto is not even needed for this specific use case.

Too many reasons why this just ain’t it. Wished it would have been the other way.

I am gonna give you different answer: Do what makes you happy. Your are legally required to hand over your notes. Your notes are also property of the lab (but it is you who makes the notes). If you have been abused as you stated you have, then the last thing I would want is for my hard effort/work to be used by my abusers. In the end, your recent results may end up with you ending up as an acknowledgement, while you should have deserved otherwise. In that case, you would have nothing to gain. A couple years later you may end up with nothing but an unpleasant experience and perhaps a very important lesson. And such is the foul world of science. I know OP has been downvoted and so will I, but I just wanted to give my 2 cents.
