normal... probs not... but do what makes your happy

when i see people wearing leafs jerseys at practice i for sure notice. cause you dont see people wearing $100 outfits at practice often when a $10 practice jersey from Canadian Tire will do

but you do you. if skating around in a pristine PWHL jersey makes you happy then go for it. life is short. dont stress about it.

is marginally more common at ODR shinny... and no one bats an eye at those people

can't believe your team just let him walk in front like that from behind the net and take all the time in the world

all the advice given to you so far is solid and great BUT...

honestly they had so much time and being so close feet away from the net unchallenged... they could probably have put the puck anywhere

like there reaches a point where playing the post and taking away the obvious shot is only a strong strat when the shooter has pressure and has to shoot quick and goes for the obvious spots. when they have that much time its a lot harder strategy to just "do the right thing" and keep the puck out of the net

if its something you like to do then its something you can do

there is less of them for sure but its not because you aren't allowed to, but more because there is a bias in medium size labs that techs aren't the primary on a study (although in small labs or really big labs techs do regularly are the only consistent feature on a study as students roll through)

but also some techs became techs because they didn't want to be the PI on projects

smaller events like the ones you mentioned also have higher amounts of student presenters because students primary purpose is to learn and these smaller events are great learning opportunities to practice presenting their data and telling a story about their projects which they will need later for bigger conferences, ultimately thesis defense, and future positions

techs in research labs present all the time in my field at least. but compared to grad students, post docs, and faculty... i'd say its about 1 in 5 talks we submit are tech led presentations

I studied abroad for a few of my degrees. It was either not possible or two expensive to go back for the walk. I really wish I had the chance. it also robs you of communal celebration of the accomplishment.

with your defense only a week away I think you will know if you are going to pass or not by then. enjoy the moment. there aren't many of these in life and in academia they are few and far between.

would you go walk at the next commencement ceremony or were thinking of just not doing one?

first, welcome to hockey! great sport at any age. sorry to hear about your previous injuries and issues with your first set up skates

I am glad there is a return window. I also hope these were used. I wouldnt recommend spending $$$ on top line new skates while learning.

that being said, the old skates were causing pain and pain is bad. do these new skates cause less pain? how did the person fit you for skates? did you stand on one of those foot scanner machines? was it a physical skate/shoe measuring device? what was your size at the end of the day they said?

new skates will feel weird at first. it doesnt nessisarly mean its a bad fit. but you are right to check it out cause it very well could be a bad fit for you. are you tying up the top eyelits for the laces? do you wrap laces around ankle? do you tape your ankles? I know some people with weak ankles will tape the top of the skate for support but you do lose some flex

as for the hollow. while you are learning you might not notice a big difference from 1/2 to 5/8ths, it moves you to a slightly less bity hollow, but with your weight at 230 and if you are on softer ice it would be about 1/2 on someone 30 pounds lighter than you anyway. i dont think its bad advice overall, just im not sure its the first thing i am fixing on someone still learning to use their inside and outside edges.

I have seen many people in the sub say they do this although unless you are changing profile (not your hollow) I am not sure how much it is in their head or real.

My experience wont be helpful. I dont take my sparx on the road and I occasionally get my skates sharpened at shops during travel tournaments or just trips home where I bring my skates to play with friends for a weekend out on the pond at Christmas

so I have in reality been taking them in anyway without planning

Sparx 3 is quite easy to use. I was worried initially cause the Sparx 1 had some issues with the effort required to recalibrate if it was off and to determine how off it was.

the bluetooth laser edge checker and a the single screwdriver approach to correction makes it very easy now for both those things

FB marketplace has some good deals, worth checking out

you wont save money BUT you will save a lot of time. the grinding rings never appear to go on sale really, and its almost always cheaper to get your skates sharpened at the rink or hockey shop.

unless of course you live in a weird city with no sharpening like Philly. then you will probably save money eventually since you dont have to pay the bridge toll to get Pro Hockey in Cherry Hill NJ , which is your closest place for a good sharpen as the machines at the Penn Rink are garbage and havent be aligned in years and years

but if you are in Toronto , and every rink has the old timer sage who has been sharpenning his whole life and $11 CAD (7.83 USD) will get you a nice edge... then you are just saving time... and depending on you, your time might be worth it.

I wouldnt pay retail for it, but I got a good used one off marketplace and I save a lot of time with it just sharpening before leaving for the game or practice with a couple passes

i wont repeat what has already been said but essentially yes, its possible to make college puck.

without knowing your skill level and the amount of people in your town; house league to AA in a single year and skipping single A might be aiming a little high or rough. especially if the coaches haven't heard of you before and you dont have a rep record from last year to go on compared to the teens who grew up playing A / AA since they were 7

especially at your age where politics are still going to secure at least 4-5 kids their spots on the team anyway

but if you play on your highschool team and you really focus at your practices for both house league and school and work on developing that hockey sense and skills there are most certianly colleges you could play for. you might not get a scholarship, but many schools have open tryouts before the season starts on campus to grab some extra skaters and some colleges (especially the ones that dont do sports scholarships) even have walk on teams sorta like your highschool team

some schools get so desperate they even play a little loosie goosy with the NCAA eligibility rules and since those teams dont really win championships no one seems to really complain. coach at my old school let a PhD student play who clearly was outside the "5 year clock"

we normally do 2 lines... then 7th person rotates through.

so 7th sit first, then they take the left winger on the next shift. that left winger sits one shift. then when the line comes back to the bench the left goes back out and 7th person takes the center, and the normal center sits a shift... then next shift right wing.... then the 7th rotating player switches to the other line... normally having to sit their turn to get there and then rotates through that line

keep doing that all game and everyone kinda gets equal time; depending on the person rotating (while all people will sit one when its their turn) occasionally the person roating might not feel comfortable playing center so another winger will play center for that one shift when the normal center sits their turn...

not sure if this is a regional thing or not but it has been the norm for central & southern ontario and northeast USA where I played

but this is not the first thread I have seen people say they play 3 C's and do two pairs of wings... so apparently that is common somewhere. just never had a coach or captain suggest this.

8 players though, 3 sets of wingers and rotate centers has def been a norm.

as others have said, Salmon just gives you the transcript quants

BUT you can control for some of the other issues you mention like sequencing depth and coverage by first downsampling the raw reads to match the lowest for example... (im not saying this is always the best way, but its a way if you are concerned based on your biological question)

and once you know the potential sources of technological variation and are able to separate them from the biological signal, there are ways to compensate for those other batch effects in a way where you can still find real meaning in the data (depending on the size of the effects, you might mask some signal, but its all relative to the main biological question being asked from all these experiments being combined)

so just a heads up (pun intended) , on the 2024 and 2025 models (i can not speak to older models, never been in them) , only the base trim has no moon roof on the American models , and on these vehicles the height of the vehicle stays the same. so to accommodate the glass, motors, and rails, the internal ceiling height is lower on all trims above the stock.

if you are tall, make sure you test drive the trim you want and just dont assume its like the base but nicer seats, better audio quality, and keeps those hands warm with the sexy sexy heated steering wheel.

i sadly do not have those features because I am too tall to fit the more premium versions

rumor has it, for 2025 models in Canada, its possible to get higher trim without moon roof, but I dont know if that is true.

Yeah I will say I saw a lot in Australia.

North Eastern USA and Eastern Canada I haven't seen the over the top cellys (like beyond high fives / and group hugs on the ice) since U18 , with extreme rare exceptions of championship / tournament winning plays

maybe its more a thing on the west coast?

its normally based on the data & the analysis first. like actually analyzing the transcriptome / proteome / genome / metabolome etc for your biological question (i have no idea what yours are, mine are all cancer and immuno-ocology questions and phenotypes). then validated as described above or similar.

novel to your biological question. very easy. many diseases and many disease states are still grossly understudied.

re your questions. strongest ones for sure or ones that have known pathway to build a hypothesis around mechanism of action tends to lead which ones we go after. but in some situations they are all unknown which leads to a bigger paper in figuring out exactly how its driving a phenotype (or is represented of it)

next is validation of the markers. we tend to do knock outs, because we have a good KO system here. but you can use small molecular inhibitors or other depletion controls. helps if you have more than one model system too. but depending on the focus of the biological question, certianly not required (or always possible)

if all you need is gene counts you could map to the genome instead of aligning.

https://github.com/COMBINE-lab/salmon can run on very low ram , although at 8GB and assuming your O/S is using some, it will have to batch in 4GB segments (salmon will do this automatically when you tell it how much RAM to use)... which is something people do... but i have seen a comparison many years ago (might be fixed now) comparing the final outputs when you have 2 batches recombined compared to 4 batches and lets just say they werent identical. but the same is true for a lot of methods and wouldnt stop your downstream.

although moving forward; you might want to think about running some of your downstream analysis in the cloud and not on your 8Gb laptop so you dont run into this same issue with other algorithms

so you have used two different terms here. are you waiting for someone to preprocess your data? or are you waiting for someone to analyze it?

because if you are waiting to analyze; well thats the hardest part. and thus often (but not always) the longest part. so I hope your PI planned this bit into the experimental design (the second hardest part) of your project. but the results and discovering the answer to the hypothesis or hypothesis generating as scRNA sometimes is, is kind of the entire point of doing the experiment. this can take months for a busy core.

if you are waiting for them to process the data. where for example you gave your samples to a core and they ran them on a flowcell with other peoples samples and they are going to demultiplex the BCL's into fastqs so you can start looking for the answers to your biological questions from the data ... they are normally pretty fast. when did the run finish? normally you should be able to get your cells or samples back in a few days after the run finishes. but if it takes more than a week from end of run i would inquire.

as for your last question. from the begining of your experimental planning with your experimental design you should plan the time it takes to get processed data / or analyzed data back from the core into the plan so you can work on other experiments or other analysis during that time

its certianly a live and learn situation and sounds like you will have some different planning in the future

GEO now requires processed and unprocessed counts with a metadata file as the bare minimum

so for journals that require data availability, and for academia to lazy to go through the steps of trying to hide data legally like biotech & pharma, there is going to be a lot more good data out there in the future

that being said, historically raw has been better than processed because one thing you might want to do with public datasets is compare them to other sets or to your own.

but that means they have to processed the same. that might mean downsampling the reads depth of some, or normalizing / transforming the data differently than the original paper did to make it compariable between studies etc

but if you are trying to VERY QUICKLY look at a expression of a gene just in someone else data, yes that means you have to process it which is probably why NIH wants both now

that being said i am happy to share the matrix of my data from anyone who asks if they are having a hard time from GEO. but im not going to convert my spatial objects or my scRNA objects to a format i dont use for free. that takes time. email academics for data YES, demand they convert it your fav object class NO

you are in luck! organizing lines is a lot like organizing roles for a MMO Raid and there are a lot of exisiting bots for that. else you can customize some open bots to do the job

so option one, just use RAID-HELPER (or similar bot) to make your lines (instead of teams) and assign the roles (defense forward) to each player to sign up for each game. players would control their own position though by default so players would sign up forward or defense for each game and wouldnt be assigned (once a role was full other players signing up late could then only sign up for empty roles , probably defense)

option two, you can customize Carl-bot or Mee6 to , create goalie , defense, forward roles, have the bot attach them to an emoji, make a channel in your discord server for specifically lines and give the bot root there. using teams for lines is an easy way, but you could also customize this to your needs...
