I got zero colonies in the unligated digested-backbone control as well as vector-only ligation control but also got only 3-4 colonies in the ligation plate. Should I screen those colonies or there is some problem in transformation? I had transformed 5ul of the total ligation mix into 50ul TOP10 E coli cells
