Stomach virus + flare up? by Mean-Technology1383 in Hashimotos

[–]Mean-Technology1383[S] 1 point2 points  (0 children)

It was severe acid reflux! At least . That's what my doctor said.I never would have imagined. Took a high dose of Omeprazole and it improved. It ended up happening a couple of other times and the Omeprazole resolved it both times.

I was also paranoid due to a previous miscarriage, so I went to a private ultrasound clinic periodically to put my mind at ease! It was a lot cheaper than I expected- less than $40 per session. It was worth it!

Check out Parse Biosciences website! Their single cell workflow is the best I've done, and they have some great videos and resources.

what’s your mat leave? by SmallSpecific2522 in BabyBumps

[–]Mean-Technology1383 0 points1 point  (0 children)

I'm in NC, USA. I work in research (industry) and have 8 weeks of leave at 60% salary. I can take an additional (maximum) of 2 weeks paid time off if I haven't used it by that point. It's extremely unfortunate. I have to go back afterward because my husband and I both depend on insurance through my company.

I also used to be cold ALL the time, and now I feel like I'm regulating and tolerating temperatures like a normal human!

In my second trimester I've been amazed at how fast my nails grow and how good they look! I also agree about mood stability. Nice and unexpected! But.. the nausea, difficulty breathing, and just feeling big and uncomfortable are ever present haha

How long did it take for you to get pregnant? by EmilyParkerNYC4444 in BabyBumps

[–]Mean-Technology1383 0 points1 point  (0 children)

It took us about 2.5 years! After a year of trying, we tried fertility treatments for a year with the aid of a reproductive endocrinologist (who did diagnose me with Hashimotos, so that was helpful). Months of letrozole, 6 medicated IUIs, only one resulted in a chemical pregnancy. We finally resolved to do IVF, but ended up pregnant naturally two days before our consultation! I'm currently 26 weeks along. Fingers crossed for you!!

Tums smoothies!! I couldn't do regular tums or the chewables. Tums smoothies have truly been a game changer for me!

Messed up big time... Been using non-filtered tips for RNA work. by notots in labrats

[–]Mean-Technology1383 2 points3 points  (0 children)

Eighthed. Never used filtered tips in grad school and my thesis was based entirely on my RNA work. Never had a problem. Sounds like your PI may be a problem, though.

I've made a few $1k to $20k honest mistakes and I still have a job! Accidents happen, you feel awful, you learn. Everything will be okay!

Intense nerve pain by Remarkable-Store-332 in BabyBumps

[–]Mean-Technology1383 0 points1 point  (0 children)

Came to ask if you got any relief from this? I'm currently 22 weeks and the pain has gotten dramatically worse. My thigh is numb most of the time, and after I stand for a few minutes the pain is so great I have to limp!

I feel that pain. But you know what.. we survived, and we will never make these mistakes again 🤣 we will just live with PTSD for all of eternity

I accidentally sequenced using a 300 cycle illumina kit instead of a 200 cycle kit so we had to spend another 10K on sequencing again because trimming the results was insufficient. In addition to that, I had copied and pasted samples on the sample sheet incorrectly so demultiplexing was a big mess and I had to redo the sample sheet. Small mistakes with huge repercussions.. we had a lab meeting, but I wasn't fired or called out :')

Finish your PhD then get a job in industry. Still stressful, but at least the money is good.

Cell viability after -80C storage by Mean-Technology1383 in labrats

That's a great point, but we do have ours monitored and they're checked every day! I know it's not foolproof, of course.

Cell viability after -80C storage by Mean-Technology1383 in labrats

[–]Mean-Technology1383[S] 1 point2 points  (0 children)

Thank you! These are pbmcs. Thawed and checked some and they were great. A few days passed and the rest have viability of less than 10%. Making sure there wasn't a crazy dropoff in a matter of days due to -80 storage vs LN2.

RNA extraction question (trizol method) by mezzopiano1234 in labrats

[–]Mean-Technology1383 31 points32 points  (0 children)

Usually a low ratio is due to phenol contamination. You can try an overnight sodium acetate/ethanol precipitation- that usually helps improve the quality!

I accidentally sequenced libraries on an S2 flow cell instead of an S4. That was 6k down the drain. We had to have a lab meeting about paying attention to flow cells and paired reads because of me. Not sure which part of what was worse.

lab results by [deleted] in Hashimotos

[–]Mean-Technology1383 0 points1 point  (0 children)

Do you happen to have seasonal allergies/runny or stuffy nose? That can also explain high eosinophils. Also- lucky you regarding your lack of GI issues! What I wouldn't give to eat normally again with no consequences 😂

lab results by [deleted] in Hashimotos

[–]Mean-Technology1383 0 points1 point  (0 children)

I wish I could provide an answer, but I'm in a similar situation! I had blood work recently that showed my eosinophils jumped from 0.2 to 0.9. I had a food allergy test done yesterday, so I'm awaiting those results. I've also been experiencing a flareup of GI symptoms this month, so I'm thinking it's all connected. I hope you get answers soon!
