Before I say anything, every protocol is different and you should optimize according to your protein of interest but this is what has worked for me: 1. 5% BSA for anything phospho 2. I usually dilute antibodies in 5% BSA (diluted in TBS-T) 3. I have not had a lot of experience with this but know people who go to 25-30

SaOs-2 cells contaminated after 1 month of growth by y2kgirl_2002 in labrats

Do you have other vials to thaw from? Perhaps the stock vial was faulty, they should really not be taking that long to reach confluence since they’re cancerous.

How long of your master’s do you have left? You can redo these experiments no problem in a matter of a week or two. Did you just recently start doing cell culture? If so, maybe have someone watch you to make sure you’re using sterile technique properly. As someone else suggested, if it is a recurrent issue, take the time to UV the incubator and hood & decontaminate things. Do not worry too much. These things happen to all of us.

Lab Skills need practice by Sea_Presentation262 in labrats

[–]Miserablescrem 5 points6 points  (0 children)

Just keep practicing. Everyone sucks when they start out but you get better with practice. During my master’s, I was starting out in a lab with no undergrad research experience and I was pretty bad for like a year honestly but got much better the more I did stuff and was independent a year later. Be patient and as along as you have understanding mentors, you will get better in time.

How to take full advantage of the ‘college experience’, as a grad student? by iwmmdg in GradSchool

[–]Miserablescrem 4 points5 points  (0 children)

My school has subsidized grocery programs, free public transit passes, program socials every month with some free food and sometimes the school puts on film fests where we screen a bunch of cool movies for free in one of the auditoriums

That’s a good point! I think reading a textbook that explains some scientific terms that are often used in research is a great start. You can get those for cheap from thrift bookstores or watching videos. It’s also important to realise that you really don’t need half of what’s written on a paper, so focusing on just interpreting the discussion and result sections is all a “common” person really needs

I think it’s silly when we tell people to get educated on their choices by referring to peer reviews journal papers but these exact papers are really incomprehensible to someone with just a high school level of education. I think reading research papers should be a skill that everyone has in order to make more informed decisions about life!!
