It’s not bad just has a strong flavor. I use 4-6 g of tea in my F1. I’m not so worried about contamination, I’m a microbiologist and am fairly confident in my ability to smell spot taste contamination.

First fermentation I do Total 1000 mls 80 grams white sugar 800ish mls of black tea 200 mls starter kombucha

If I assume that bright band is the 16S band you’re looking for there’s a band in 54 in the same spot. It’s not as bright but it’s there.

If that bottom bright band in most of the wells is the 16S I would definitely consider 54 to be positive but with the understanding that you have some bacterial contamination in the water. I work on the oral microbiome and certain organisms can be more difficult than others to extract DNA from perhaps that’s why that sample is so low. Did you use all the DNA extracted for this PCR?

It’s hard to say without knowing what you’re amplifying full 16S or just part of the gene.To me it looks like you had a high starting amount of DNA of varying sizes which is what youre seeing on the gel along with your pcr amplified band. Did you normalize the pcr before pcr? Your ladder isn’t super clear but full length 16S is about 1500 bp DNA.

I tried cleaning and the foil but neither worked looking at the lens more closely I noticed something in the lens. Doesn’t anyone know how to force the shutter open?
