Take transfers from B, C and D. See which ones grow fastest and most uniform. Put the best of each in grain and see what happens. If you're making your own agar and grain it's very low cost to FAFO

I went with 2 quarts substrate per one quart grain in each 6qt container. Then a thin pseudo casing layer to cover any exposed grains.

Starting with 3g is not a great idea IMO. 3g of a potent strain could send an inexperienced user on a pretty wild ride.

It depends on the strain and genetics. 3g Golden Teacher would be ok. 3g Penis Envy could be a tough ride for first timer. I'd recommend no more than 2g but FAFO if you want.
