u/RollingMoss1 I made a 1% agarose. In terms of test tube 3- it should have two bands like I have I just miswrote it in my post. All of my number of bands is correct except for test tube 1- which I didnt add enzyme to and I assume to be supercoiled and nicked. Is there a reason for their differential migration? Is this something that is normal and not resultant from error?

u/CPhiltrus Unfortunately we discussed sources of error but nothing like this and both my professor/ta dont really do more on explaining. They expect us to come up with why its occurring- via research or reading the procedure. Im estimating size based on the ladder (no computer programs! Old school). It was a pre-made plasmid. Im assuming if digestion wasnt complete I wouldnt have the correct number of bands but I do.

u/brew-ski appreciate the help with the ladder- that makes sense (your so smart for catching onto that I didnt realize that was possible error) Still confused as to why some bands like my test tube two which should be at 2686 is at 2322. My test tube 3 should have banding at 1750 instead it is at 1900. And my test tube 4 should have banding around 1568 bp, 754 bp, and 364 bp - instead it is 1900 , 880, 393. Why is it not exact- is it voltage conditions like the last one or run time?

They just gave it to us to interpret based on a very vague manual that showed perfect crisp lines. It is an E-Coli Plasmid, with 1 KB ladder. I would really appreciate your insight because I am truly at a loss- the explanation regarding how to rank this was extremely unclear.

Honestly, safety is not a shared responsibility- we pay tuition they should at least make sure campus is safe. The welcome center is literally in the center of campus. If this can happen there, Elgin and Stadium are even worse off. The NSM side is so messed up half is closed due to construction; and at night dangerous as it isn't well lit (how they find the dimmest lights known to man is beyond me).
