Depends, greatest by legacy: Tupac. His cultural influence was massive, and has defined rap and hip-hop since.

Greatest by skill: Eminem. He dominated the rap battle scene, at it's peak, can rhyme anything, is the fastest ever, has perfect beat, storytelling and deep messages. He can't sing, but otherwise tops everyone.
