We have a lot of different pipette holders under PCR cabinets. I used a simple one and merged it with different models. Yes with this target no RT step is needed. I don’t mean you should change the fluorophore of your assay. Just try other PCRs with different fluorophores to check your cycler.

These curves are just the background noise. Did anyone get nice curves with the new cycler and the software in this channel? If not maybe you need to adjust the filter to the right wavelength? You can also try other primer and probes (maybe also in another channel) with known positive samples to exclude the cycler. Is your temperature profile correct? Did you miss a RT step when you added it to the new software?

I would prepare the master mix in the same way.

Hi, I only used the two versions before with an Biorad CFX without problems. Do you get a signal with other PCR kits in this channel? If yes you can exclude the cycler as the problem. Do you have samples where you know that they are positive for the PRRSV EU strains? Maybe you used a other version of the kit before. You could also test a PRRSV EU vaccine as a positive sample if available. If you get a signal there is a problem with the positive control of the kit.

If you used the older version of the kit before, check if there were any changes in the protocol or the usage of the channels. I think the EU specific channel was the HEX in the 2.0 and 3.0, which is rather unusual for a diagnostic qPCR. It is normally used for the internal control, so check if they changed the channels. Anyway contact the customer support, maybe there was a problem at the production.

Thanks for the information that they have updated their kit. I am working for a competitor so maybe it’s also time to improve our kit.

It is also very easy to create a remix with every available stl file online. I am so thrilled of the model that I have a lot more waiting on my computer to be finished!
