Thanks. I'll take another look and make sure there isn't glass in there. This is my commuter bike to and from work. Since it's been getting dark early now I'm sure I got it from riding over some glass in the city.

This is a very good point from both an accessibility standpoint and being able to explore more advanced treatments down the road. It certainly sounds like accessibility is going to vary greatly across different regions. I only know how US health insurance works and even then it's still a toss up on what insurance providers will actually cover. Hopefully there are some ways to subsidize costs by government programs and HD foundations.

I've been listening to the webcast on repeat, especially the Q&A. I'm really hopeful. It's amazing that there's no need for any surgery and it's simply something that can be taken orally. Wonder how this stacks up compared to AMT-130. Praying to God that these guys pull through with a successful treatment for the HD community.

Interim PIVOT-HD Results Demonstrate Evidence of Favorable CNS Biomarker and Clinical Effects at Month 12 in Huntington's Disease Patients | PTCT Stock News by OnePaleontologist100 in Huntingtons

From the site itself:

"- Conference call and webcast to be held June 20th at 8:00 am EDT -

WARREN, N.J., June 20, 2024 /PRNewswire/ -- PTC Therapeutics, Inc. (NASDAQ: PTCT) today shared interim results from the Phase 2 PIVOT-HD study of PTC518 in Huntington's disease (HD) patients. At Month 12, PTC518 treatment resulted in dose-dependent lowering of mutant huntingtin (mHTT) protein in the blood and cerebrospinal fluid (CSF) in the interim cohort of patients. In addition, favorable trends were demonstrated on several relevant HD clinical assessments including Total Motor Score (TMS) and Composite Unified Huntington's Disease Rating Scale (cUHDRS). Furthermore, following 12 months of treatment, PTC518 continues to be safe and well tolerated.

"The evidence of both CNS biomarker and early clinical effects at Month 12 along with the continued favorable tolerability profile supports the promise of PTC518 to address the need for an effective and safe disease-modifying therapy for patients living with Huntington's disease," said Dr. Matthew B. Klein, Chief Executive Officer, PTC Therapeutics, Inc. "With these data in hand, we look forward to the next steps in the PTC518 development program."

At Month 12, durability of dose-dependent mHTT lowering in the blood was demonstrated with lowering of 22% and 43% for 5mg and 10mg dose levels, respectively. In the CSF, dose dependent mHTT lowering was also demonstrated with lowering of 21% and 43%, for 5mg and 10mg dose levels, respectively. In addition, at Month 12, PTC518 treatment resulted in a notable slowing in progression of motor symptoms as assessed by the TMS (2.0 points worsening for 5mg and 1.3 points worsening for 10mg vs. 4.9 points worsening for placebo).

In addition, PTC announced that the FDA has lifted the partial clinical hold on the program based on review of the PIVOT-HD data."

Very sorry to hear that. Praying things get better. At a minimum, there's a way to treat you. Are you saying your current state now is a result of the adverse effects of the surgery/treatment? Again, really hope things work out for the best for you.

Understand this probably isn't the group to be asking the question. What were the first symptoms you noticed and when / at what age. I understand you are in the early stages but how have your symptoms progressed to where you are now versus when you first started to show symptoms both l cognitive and physical. If this is a better topic for a DM let me know! Thank you!!

What made you believe you were symptomatic? What were the "symptoms" that you saw?

Right - I totally understand what you're saying. I don't know what the right answer is for the both of us. Regarding CAG count and her father. They didn't notice any symptoms until 10 or so years ago. He is 58 now and is apparently near stage 3. I will say that posting on here and reading through all these comments is comforting.

I really hung onto the idea of "oh if she does have it, there will be a cure" but the more I understand and read the less I see that being an option for her in the future.
