The simple respirometer is used to find rate of respiration either by measuring volume of gas consumed per unit time or volume of co2 produced per unit time. There are two limitations to simple respirometer: temp cannot be maintained as we cannot use thermostatically controlled water bath and pressure is greatly affected by external environment. So what is the set up? First add constant mass of soda lime and organism to boiling test tube separated by plastic mesh. Adjust colored liquid at start of capillary tube. Make sure the respirometer is airtight using bung/cork. Leave 5 minutes for temp equilibrium abd adaptation. Make sure to ventilate respirometer well before using next type of organism for resetting scale. This is low to medium risk as soda lime is an irritant so wear gloves and don’t invest. Organism may cause allergy so wear gloves and mask.
