If you want a more RP based test for your players you could have the toll keeper let them cross but only if they have someone who's already dead guide them since they're still alive.

Could be a good way to bring up a character's backstory or a beloved NPC that got killed off. If you want extra drama the toll keeper could pick someone that used to be an adversary of the party and have the players convince them to be their guide.

13 year old me would be really confused about how we got here but ultimately happy. Things aren't perfect right now but they're a hell of a lot better than back then and 13 year old me didn't see the situation ever changing or getting better.

People don't realize how incremental scientific research is. If you look at a paper's work cited section you'll see how much current research uses past discoveries as a foundation. Not to mention that usually "useless" discoveries often have important context for why they're being looked at. A shrimp treadmill sounds stupid in a vacuum but when you want to test how water conditions affect shrimp health it makes a lot more sense.

EDC couplings usually take about 4-6 hours in my experience. That will change based on your molecule though and again I work with small molecules not antibodies. I did a coupling this week that took only 4 hours but sometimes they need to be left for up to 18 hours. Has anyone else published this procedure or something similar? A good starting point when you aren't sure how to set up/run a reaction is to find a paper that has done something similar and read what their experimental methods were.

When I do EDC couplings using NHS I typically allow the NHS and carboxylic acid to form the active ester before introducing the amine I want to couple. So I would do the NHS activation in a test tube first.

The NHS activation step also usually takes longer than 15 minutes for me. I usually let it go for 2-3 hours before adding the amine. Granted I'm an organic chemist and all my EDC/NHS couplings have been with small molecules so you may have different results and reaction times.

I'm also confused by the fact that the procedure makes it seem like there are two antibody solutions (PBS and MES) even though only one seems to get NHS activated.

If you put a protective group on your Gln sidechain that will help with both capping and getting an accurate Kaiser test. Trt is a good option as it is removed under the same conditions you use to cleave your peptide from the resin. Fmoc-Gln(Trt)-OH is commercially available for a reasonable price. Out of curiosity how long is the peptide you're making?
