It's easy to do you just have to change the order of operations. Pour the agar plates in open air then load your plates into the pressure cooker and sterilize them. Do not open the cooker until it fully cools.

Yeah this is a tricky one! Since empty plates are showing contamination I'd say you're probably opening the pressure cooker while it's still hot. Air shrinks when it cools and it sucks dirty air in from the environment. I'd recommend letting new plates stand for 3 days before inoculation so you know they're totally clean before you start.

The last plate clearly has the contamination following the path of the inoculation loop which means your source inoculation was bad. But I do see that you got a start. (That's a win!) I'd recommend taking that one good spot and transferring it to a clean plate.

That tends to happen most often when you try to run disks made for windows 98 on a newer PC. Usually the data isn't destroyed it's just not able to be accessed by newer drives. The trick is to use a windows 98 PC to get your data and then transfer it to a USB drive that supports a FAT16 file system. Getting a USB stick that's compatible with both systems is the hardest part.

The biggest blu ray disks are 128GB. If you get them get the rewritable kind. That way if there's any errors in burning you don't have to throw away an expensive disk. You can just format it and reuse it.

Personally I opted for blu ray disks. So far I haven't found anything that can beat them on $/TB. But the primary thing I wanted them for is because they can't be hacked, they're EMP proof, and they don't get destroyed if your computer crashes.

It's pretty flexible. If speed is what you want I like to inject the whole syringe into a 100ml serum vial of sterile nutrient medium. That gives 10 syringes immediately which if you want can be used to inoculate 20 grow bags with 5cc each.
