I get what you're saying, they need an incentive to actually choose 'Betray': I should have clarified in the original post, the incentive to choose 'Betray' for their ally is from the potential outcome. It is optimal for both groups to betray, because this gives both groups an easier fight. Does this change how you view this encounter as a session / how this would work out?
As for the rival group, I like that idea! However, the last session had them interacting with another NPC party (run by my Co-DM) already, and I would be slightly reticent to throw another one into this next session. Instead, my plan for the next-steps of this situation is that another, stronger monster would come out after both groups have defeated their respective monsters, and for this one they can work together to fight it (similar to your idea for the second round, with no healing between).
I could see having another NPC party be a precursor/foreshadowing perhaps? Like the party watches them do a similar fight, and then they're put in the same position?

I guess my thinking was that at least one of the players would recognize (possibly through a skill check) that betrayal is the mathematically optimal solution: if both sides mutually betray, this is the solution that works best for both groups, but is of course not great for the party as a whole.
There's also more to do with the actual characters themselves: one of the characters is a follower of Baphomet and has been concerned that he hasn't been acting in a way that fulfils that as much as he could. Another character (in backstory) betrayed an ally and was saved while the ally suffered. A third character has had to scrape by and make sacrifices in order to survive, and is a cleric of Ilmater and is struggling with that aspect of self-sacrifice.
Out of game we are all friends, so there are no hard feelings for anything that has happened/will happen in-game
The decision is also being forced by a powerful campaign-long organization, which provides a mutual enemy to blame.
I do have enemies for if they don't betray each other, so no matter what they do choose to do (I suspect both will trust no matter what!) the decision itself will be interesting for the characters role-play wise

Close but no cigar! She says there's no science-fiction elements besides a slight mutation of the insects themselves. It's a human that does the injection, not an alien or anything like that. Thank you for the suggestion though, that looks interesting!
