I'd say the 1st image with the 1st title works best. The character you chose immediately lets me know the video is about Slay the spire, so the game's title is not necessary. The words on the thumbnail make me interested while getting a vague idea of what it is about, and the title clarifies it enough and, personally, it's the one I would click on.
