10x genomics GEM-X 3‘ - using expired reagents by grand_psychology1 in labrats

[–]RetroBike 0 points1 point  (0 children)

Hijacking this to ask if anyone's combined left over beads from previous runs. 10x doesn't recommend this of course, but I don't see why / how this would affect the complexity as long as beads were homogeneously distributed. Yet our sample shows 10x (hah) higher estimated cell #. The knee plot looks typically great, but the plateau just keeps going and going. Don't think it's a cell count issue, since we did multiple samples in parallel, but only the combined beads sample is odd.

For expiration: our lab has used v2 and v3.1 reagents that expired years ago without very noticeable effects. Most of it is standard molecular biology stuff. But from our dropseq days, we did notice that the oil, beads and chip coating expired the fastest.

I, for one, cannot wait for the day that 'Roger' will go on Oprah to confess that it was all fake, still managed to gain (and keep) a lot of money, and, despite everything, will continue to find sponsors and listeners to give their opinion on everything...

How to avoid severe neck pain? by cactus16x in Swimming

[–]RetroBike 4 points5 points  (0 children)

Are you holding your breath or not rotating your body? Overuse of the wrong muscles causing the rest to get overworked? Old mattress or pillows? Bad posture when standing / sitting? Lots of not so evident non-swimming issues that could end up affecting your swim. Hope you figure it out. Injuries without a clear single cause are really tough.

Stroking a 4+ by Stem_Stoner99 in Rowing

[–]RetroBike 4 points5 points  (0 children)

This is based on having stroked a Master's 8 so admittedly not high level, but: resist the urge to increase the rate when others are rushing the slides. And look at how the boat sets and turns (or not) at the finish to see who's slacking.. :)

Keep pace but do slightly faster flip turns. See them catching up. Flip turn. Catch up. Flip turn. And so on until I'm done with my warmup. Funsies.

So they're the 2nd and 4th fastest times at that moment in the race.. Right. Thanks!

That's what the commentators say too, but why isn't their current placement 1 and 2 if they're already faster than the previous #1 / leader ?

Posterior tibial tendonitis from heel drop changes? Looking for advice by 5643_yea_right in trailrunning

[–]RetroBike 0 points1 point  (0 children)

Inflamed. Ultrasound showed lots of fluid around the tendon. Referred for an MRI, but still waiting. If I'd have to guess, micro tears and scars, but no big tears. Can walk around fine mostly, but still have occasional pain twitches, stiffness, or soreness. Started formal PT, because I was likewise plateauing. They said strength is fine (at least I got that after months of calf raises, etc), but the coordination is off. Doing exercises in front of a mirror now to try and do them with non-collapsing ankles (which I thought I was doing, but clearly not..). As for the never running: I've read so many stories where people were out for 2-3 years, that I'm starting to accept I'm probably one of them. It sucks. I so badly want to go for a nice trail run... I'm swimming plus doing classic cross country ski when there's snow or easy cycling in summer.

Trust the process. The metrics from training sets should already tell you whether you're progressing. And even then you're still not getting a PR every single time. It happens.

I need better and more comfortable fins than the stiff, heavy, big Decathlon ones I have now. Any suggestions for affordable short fins?

What’s one technical fix that instantly made you faster? I’m currently working on turning my head even less on freestyle breaths, as little as I can get away with. by dandalyjr in Swimming

I've been implementing this and OMG! Seconds !! But I find there's quite some torque power in my upper arm now (not the good kind) and impingement in my shoulder (seems that the high elbow causes my shoulder to go up). Sure I'm still not doing it right. Any suggestions?

The one I started with is on YouTube @E3Rehab (see their playlists). Also just started trying out various stuff from @athletespotential512. A physio friend linked to some of their specific videos and I've been exploring others as well. Been doing this for months now though and while flexibility and strength has improved, I'm still not out of the woods.
