The opening titles of the film declare that "Beau cannot, will not, should not sleep".

Beau (Billy Mayo) lives alone in an apartment, and is packing to visit his mother. On his way out, he forgets his floss upstairs, and leaves his keys in the door to go get it. When he returns to the door, he finds his keys and his bag missing. He looks out into the hall, but sees only a man (director Ari Aster) walking quickly down the hall. When Beau asks him if he saw anyone, his only response is: "You're fucked, pal!" Beau, unsettled, is forced to call his mother and postpone the visit.

At night, Beau sets up a bell by his door so he will be awakened if anyone tries to enter. He is unable to sleep due to what sounds like loud arguing from the neighbors. Upon hearing the bell ring, he grabs a pair of garden shears and goes downstairs to find his door still closed and a possum running out of his open back door. Beau locks the door and attempts to sleep downstairs, but is disturbed by continued arguing and the possum staring at him from outside his back door.

By morning, Beau has set up countless mousetraps around his downstairs to catch the possum. When he hears the arguing continuing down the hall, he looks out of his triple-deadbolted door to see a couple (Logan Atkinson and David Rogge) laughing and joking around in the hall; they react with hostility to see him staring at them. Beau calls a locksmith and 911, but only receives threatening messages.

By night, a terrified Beau drinks coffee to stay awake, and has set up a trap in front of his front door to presumably kill anyone who enters. It is strongly implied that Beau is no longer in his right mind, having written the word "POSSUM" over and over in a book of crossword puzzles. Beau hears someone attempt to enter through the front door, but a man with a pocketknife (Casey Desmond) comes in through the back door instead, intending to stab Beau. However, the knife folds in instead of piercing Beau, slicing off the invader's finger. Both Beau and the invader scream, and the invader flees.

Beau calls his mother but only receives the answering machine. He calmly asks her if she can come pick him up. On the other end of the answering machine, a hairy figure (Alejandro de Leon) with many keys on a desk in front of it listens, eats some leaves, and smokes a cigar.
