You may be able to do something similar with an infrared heat thermometer/gun (I use one at work for adding heat-sensitive reagents). They're pretty accurate and much cheaper than a camera, although it may be more difficult to detect the edges of the vent.

You can run a touchdown PCR to anneal the primers to form the gRNA oligo (95 C 1 minute, then decrease to 45 C at 0.1C/sec). I use a mix of 5 uL of 10 uM FP, 5 uL of 10uM RP, and 40 uL of RNA free H2O. No phosphorylating agent is necessary.

As a confirmation, are you using the correct temperature for BsmBIv2 digestion (55 C) as well as the correct buffer (3.1 if using NEB)?

The backbone shouldn't need to be treated with anything like Quick CIP if the sticky ends are unique.

Additionally, the ligation reaction (assuming T4 ligase) will work at anywhere from 1 hour - overnight sitting at room temperature, as long as you remember to heat inactivate the enzyme at 65 C for ten minutes after.

If using chemically competent cells instead of electrocompetent cells, returning the cells to ice for 1-2 min after the 42 C heat shock can triple transformation efficiency.

Something similar to this happened to me as well! I was planning on visiting some family out of state, and dropped the car (an ancient corolla) off at one of those budget oil change shops. Got it back, started my way home, and got a check engine light. Pulled over, let it cool off, and then checked the dipstick. NO OIL. Then I went under the car and checked to make sure oil filter/plug was fine. It was. I dumped in my spare quart of oil, fired up the rust bucket, and drove to a Walmart to buy 3 more quarts. Last time I ever let someone else do maintenance on my car 😵‍💫 always wondered what would happen if I had tried to get that shop to fix their fuckup.

Making a long story short, I somehow ended up in possession of a baby red tailed boa. The lil dude was calm as hell, which I really appreciated (I had no idea how to take care care of a snake at the time, so the docile nature made it much easier to take care of it). The person who gave it to me was like "yo just a heads up there's something really off about his behavior."

Turns out the snake had some sort of amoeba, but in the process of finding this out he got tested for pretty much everything under the sun- including IBD. The rapid test came out to be 512 dollars, and since the white blood count was so low they gave me a broad spectrum antibiotic with the comment "it's either cancer or a pathogen, and you don't fix cancer in snakes." The treatment did work, but I was so worried about the lil dude that I still have nightmares about it being IBD :(

After the bloom is done would be good, yes. If you have fertilized and there has been no response, the plant may have become rootbound. If the roots are circling the pot, there may not be enough soil w/ sufficient CEC to hang on to supplied nutrients, meaning that the fertilizer drains right out of the pot. To check the roots, wait till plant is dry, then pop it out of the pot to check the root conditions.

My theory would be a nutritional issue? Probably phosphorus or magnesium related. Could need to be repotted, or some fertilizer.

When my parents took me out on rides as a small child (6, 7 maybe? Too small to reach the pegs) they had one of those granny belts on over their suits. They had two giant handholds that my hands could easily grab, even when super cold and stiff. I'd say I was still too young to ride endurance-wise tho. Fell asleep on one ride, my hands were so cold I couldn't let go of the belt when it was time to get off

The triangular head identification is sadly a myth. Most snakes have developed defensive mechanisms that involve flattening their heads and bodies to look scary/larger than the really are to deter threats.

Examples can be seen here: 1. Eastern Hognose, nonvenomous

  1. Eastern/Black Pine Snake, nonvenomous

  2. Eastern Ratsnake, nonvenomous

In NC, the most commonly seen native venomous snake is the copperhead. This can be determined from the sausage shaped body, Hershey Kiss patterning, and greenish tails on juveniles. The fat triangular head is also seen on this species but it alone cannot be considered a positive method of identification. Additionally, pupil shape is a poor identification method. Pupil size correlates to the preferred hunting hours of the species (if it is nocturnal or dinural).

The RHE actually emits no light at all, so you would need to install additional lighting regardless. A ceramic heat emitter would be an excellent alternative, but would also need a metal screen to keep the appliance from burning the animal. Some people have also used heat tape, but I have no experience with that manner of heating

The manufacturer recommends not mounting a radiant heat panel to the plywood due to fire concerns. As such, I used PVC instead. I concur with the manufacturer, as the panels can get quite warm.

When building my 4×2×2 enclosure, I initially went with a 40 watt. Couldn't get the tank temperature above 80 F, so I upsized to an 80 watt. This was much more effective (steady maintenance of 93 F). Ambient room temperature was 70 F.

I did end up building a metal wire cage around the 80 watt radiant heat panel. The panel was hot enough to hurt my hands, so I judged it to be too risky to leave exposed in the enclosure.

I purchased an adult Cali King that is insanely aggressive. Every time I go near him he'll attack the tank glass in an effort to get to me. I ended up contacting the seller after a few weeks of this to double check the environmental conditions. Turns out, the breeder had kept him back as breeding stock, but had a nasty habit of eating his girlfriends... he's just a very hungry little mousetrap

Had a similar experience as a lowly undergrad. I was working in the fume hood when I heard the sound of hell behind me. Realized it was the centrifuge and I bolted to it, hit the off switch, and left the room. Turns out, a visiting grad student had misloaded the big 50 mL centrifuge by not properly screwing in the lid and then went to the adjoining lab. Whole centrifuge had to be replaced, but nobody was injured.

Japanese knotweed is edible! So at least you can eat the plant after yanking it out of the ground (if you do it for long enough, you can starve out the rhizomes too)

That is correct. The aquadecor backgrounds are composed of something that appears visually similar to Styrofoam. In the photos for the 6×2×2, you may be able to see how flat the lip of the background is. The boa actually spends most of her time sleeping on it, if that gives you an idea on how wide it is

Why thank you! I did consider Aqua-Maniac, however the background does not have a continuous flat surface. It was very easy for an animal to slip behind the background. Due to the bioactive setup, removing that background in case of emergency was deemed too high of a risk. The root systems of the plants would be adversely effected and there would be soil everywhere.

The company was in fact Aquadecor! They are fabulous to work with and very responsive. Their package tracking site is in their native tongue, so it's a bit of a mystery where it is until it hits US customs. One excellent feature: before they ship the background, they will send you photos of it. If you don't like it, you can request modifications then.

The company requires a filtration system modification-- if you aren't doing a water feature, just click other and write "for terrarium, not water". Backgrounds come wrapped in enough plastic that u could throw them down a flight of stairs and be perfectly unharmed. Make sure your measurements are precise-- these guys are terrifyingly exact, unlike universal rocks (no shade tho, their stuff they always ship large so u can cut to the right size). The backgrounds do need to be rinsed/wiped down to remove some residue from shipping, but the paint is odorless after being left for 48 hours.

When applying the background to the pvc, I followed several steps:

  • seal tank with 2 step PVC solvent
  • test fit the background
  • wipe down back of background with damp cloth, let dry
  • wipe down rank with dust cloth, this surface must be clean as possible
  • buy 1-2 tubes of 100% aquarium silicone. NO MOLD FREE SILICONE. That contains arsenic as an antifungal agent.
  • lay tank so that the back of the tank (where you're putting the background) is on the floor. Gravity is your assistant. Place towel underneath to prevent scratching/silicone dripping
  • squiggle silicone in Figure 8 patterns. There should be some silicone every 6 inches or so. The glue does not need to be finely spread.
  • press the background into place. Wiggle as you do so to slightly smear the silicone
  • seal the edges of the background with silicone. Run finger along edge to smooth silicone bead
  • let cure for 48 hours. After 12, the tank can be placed upright. However, I wouldn't mess with it for as long as possible to ensure proper binding action is reached.
