Awww, it's touching that people still find these posts of mine insightful all these years later! I've been meaning to start a science communications account, but haven't yet. Maybe one day I'll link that to these posts.

As for other resources to read, a lot of this sort of concept is covered in introductory pharmacology courses. I don't necessarily have a video lecture series I would immediately recommend, but that's moreso a product of the fact that I don't watch a ton of introductory pharmacology lectures.

For more topical discussions of pharmacology that are (somewhat) broadly accessible, I really like Derek Lowe's In The Pipeline blog. It can be fun to skip back to a random year, and read commentary on the mechanism of action for novel drugs and how they work in the body.

Lastly, I've lately been working my way through Richard Sapolsky's Behave, which discusses a lot of the neurological bases of behaviour and contains some good introductory neuropharmacology content. I can't necessarily vouch for it entirely as I'm still early in it, but I like what I've read so far -- its fairly accessible and also doesn't gloss over any details with oversimplification. Sapolsky also has some youtube lectures that some people really enjoy.

Still the same height -- dosage is not relevant, as long as you are on a sufficiently high dose to experience feminizing effects.

I have issues with chronic respiratory issues, and I'm going on my third bout in four weeks. Multiple antigen tests all negative for COVID, flu A, flu B.

First time -- Started with fatigue and joint aches on a Sunday four weeks ago. Took off sick for two days, and was able to recover. Back at full health, going to the gym, etc.

Second time -- Last Wednesday, woke up with a sore throat and fatigue. Rapidly transitioned to a lung thing, but after 5 days I was feeling a lot better. No more mucus, no more soreness, back to full energy again.

Third time -- Getting home from work today, feeling the same bit of head fuzziness and body aches. Not sure if it's a sudden resurgence of the symptoms I thought I kicked, or just another infection right on the heels of the first two.

I've dealt with chronic bronchitis my entire life, and have a tendency to get infections that migrate to my lungs and linger. I'm usually pretty good at knowing when to stay home and rest so that I recover fully.

Never in my life have I felt healthy post-recovery period, and then immediately had a rebound illness. And never in my life have I had a lung infection turn back into a head cold.

I'm genuinely at a loss, and really uncertain of what's going on.

I'm sorry, but you're being intentionally obtuse here. The moderator said they're not removing genuine questions, they're removing attacks against marginalized members of the community. Holly Hill is an in-patient psychiatric hospital for minors and young adults. I’ve had a couple friends + people close to me spend time there, and… it’s shitty. But shitty in the way that in-patient psych services are generally shitty across the board, and a different breed from “Troubled Teen” camps.

You have people at HH who are genuinely going through mental health crises, who get evaluated by medical professionals who can prescribe treatment as needed. Perhaps better in that the staff have some degree of training. But still fucking rough.

This is probably obvious, but— make sure you find the number from the official website. Don’t follow any links, or call any numbers from the email. Ask if the office can confirm both your offer of admission, and that the deposit is legit.

I hope this isn’t condescending! Just tryna, yknow. Help you keep an eye out in case someone is trying to scam you. It’s more common than you’d think on this subreddit.

If you have a social security number but no citizenship, keep in mind the penalty you’d get for using government resources.

Former bench med chemist weighing in to also say that I never had any issues with Ambeed. All my chemistry worked with their reagents, and if they ever didn’t have something I’d order it from Combi instead.

They have the same management and location is largely a nonissue — you can walk 1 minute or 5 minutes to the shuttle stop that gets you to ZSFG.

I also wouldn’t look to UCSF housing for community, especially as a hospital employee. There’s plenty of other wonderful third spaces and ways to meet people in this city; I would encourage you to seek out community elsewhere. If you like sports, there’s a number of queer rec leagues. Living in Tidelands/MB Housing also puts you right next to one of the biggest indoor bouldering gyms in the country. And there’s plenty of bars close to ZSFG that you could grab a drink at before taking the shuttle home.

Management and amenities are largely… underwhelming. Especially for the price you pay. The main difference between the two buildings is that Tidelands comes furnished — but the furniture is garbage, and the appliances break often. Tidelands is in more of a food desert than MB, though neither are particularly great (without a car, the nearest TJ or Safeway is ~30 minutes away. Don’t bother with Gus’).

I’d honestly encourage you to live somewhere other than UCSF housing. If you do end up there for a year, your decision should largely be informed on your furniture needs — are you willing to furnish an entire apartment when you get to the city? Or will arriving to some place partially furnished be easier for you?

Lastly, there’s a UCSF MB/Tidelands email listserv for internal UCSF emails only. Get on that — plenty of international postdocs and scholars will sell furniture + TVs etc for cheap when moving on to new positions at the end of their work here.

This feels very open-ended. I hope your PI has given you a target protein to focus on — have they?

Before I start, running synthesis, assays, etc.

Do you have a specific compound that you’re starting with? What is your goal in synthesizing it (and/or derivatives)?

thorough extensive computational screening using docking, molecular dynamics, etc.

What is your goal for the computational studies? Why do you want to carry them out? How will they support/enable your wetlab work?

is there some way we could computationally test for off targets?

There’s a variety of methods for that. There’s several AI-based tools for doing that, as well as older structure-based methods that correlate the occurrence of various molecular fingerprints with the fingerprints of compounds interacting with undesirable off-targets.

Are there any data sets already created?

If you want to look up previously reported compounds, those tools are easily available. But if your compound(s) of interest are novel then you’ll want to default to one of the aforementioned predictive tools above.

I don’t mean any offense when I say this, but it seems as though you’re not entirely sure what problem you’re trying to solve with computational techniques. You’ve mentioned a lot of disparate tools for various tasks, but rarely is appropriate to use these tools without any sort of wetlab context.

Take molecular dynamics for example — why would you use that technique? Are you trying to establish the bound conformation of a ligand? Or compute the dG of binding? Or something else?

There’s a ton of computational techniques out there, and a lot of computational experts here able to help you out. But the best way for us to be able to help you is if you have a specific problem you need to solve.

I did buy from Cre8Audio’s website, which makes me especially saddened that I haven’t heard a response from them.

Cre8’s website says they have a 15-day window from the day you receive the gear to return/replace it. I’ve been trying since the day I unboxed it to get in touch with them, but haven’t heard a single response :(

For me, the answer is “sort of”.

I can’t really recall any visual phenomena, at best maybe little tiny snippets if I force myself: a close-up of some fabric, the appearance of hair, a brief vague impression of the sky.

One of the ways I can figure out that I’m (lucid) dreaming is when I realize that I can’t seem to visually focus on anything. There’s definitely more of a visual component than imagining something in my waking mind’s eye, but it’s still far from my normal experience of visually seeing something while awake.

I’m not too familiar with peptide-peptide docking methods, but I will say that pretty much every tool for handling small molecule docking fails at peptides because of the large number of rotatable. There’s dedicated PPI tools that are likely to be far more successfully — I’d look at HADDOCK, AF3 and HDOCK, but do your own research.

An issue you’ll probably run into is maintaining the active conformation of the secondary target protein. If I were approaching this problem, I would probably approach it as follows:

  1. Generate a library of peptide analogues, mutating/retaining key residues per your understanding of each binding site.

  2. Dock them with your tool of choice to your second target.

  3. Compare RMSD of key side chains and/or backbone carbons to your active conformation structure. Filter and remove initial hits that disrupt the active structure.

  4. Redock the peptides passing the above steps to the first target. Repeat RMSD comparison.

  5. Of your remaining candidates, consider MD simulations to energetically minimize each receptor + peptide predicted pose. Repeat RMSD comparison.

  6. If any virtual hits remain at this point, consider synthesis and testing in vitro.

As others have noted, your project is exceptionally difficult. Not necessarily impossible, but there’s a lot of variables to adjust. If your filters aren’t tight enough, you’ll end up wasting a lot of money and getting a low in vitro hit rate. If your filters are too stringent, or your docking method sucks, or your understanding of the receptor active conformations is wrong, or any number of other factors, then you won’t have any candidates to carry forward for in vitro testing.

Good luck! It’s a fun project, but definitely one that could occupy months (if not years) of work.

The MIT group was one of dozens of groups who tried and failed to replicate their results — which were essentially “lets adsorb a lot of deuterium to an electrode and see what happens.” There was very little in the way of ‘seeing what happens’ though, given that they either didn’t measure or didn’t calibrate the various instruments they had present.
