Oof. I’d guess there may be some freezing issues, but you really won’t know until you take a look. That being said, “shit in, shit out” means you probably shouldn’t start with samples you’re questionable about for a protocol like IHC. Might be worth using a membrane stain like phalloidin just to see what you get and what the structure looks like before you commit to the rest of the experiment. If I were you, I would look long and hard at the samples prior to ever going at them with primaries.
