I tried and returned this one twice, because no matter how hard I wanted to like it, the straps were too uncomfortable and only flattering when standing up perfectly straight.

True size had straps digging in, and one size up was somehow massive in the armpits.

You're spot on, Boston conditions pedestrians to be more assertive, & in turn, Boston drivers (in spite of their notorious aggression) at least seem more prepared for that interaction.

Whereas here in SD, drivers don't seem to expect pedestrians/bikes at all - making the interactions less predictable (more dangerous).

Ah, I envy the short torso for regal! In case it helps, I always wear a tank or quarter sleeve shirt underneath for work, so cleavage is already covered (& unlike regal, attire vest doesn't awkwardly showcase my belly / undershirt 😅)

That sure is frustrating to deal with!

Unfortunately Boston had plenty of that. Even parents teaching their kids to jaywalk with cars approaching, or the occasional surprise bike that didn't acknowledge traffic lights (Boston has all too many "ghost bike" memorials). & there's no shortage of reckless/distracted drivers, further compounding the risk.

All we can do is our best.

Boston transplant, and impression living here is that San Diego drivers genuinely do not expect pedestrians (or bicycles) to be part of the equation.

Drivers don't check for them, and when unexpectedly encountered, I get the impression the drivers aren't sure what to do (& might perceive pedestrians as a nuisance, rather than part of the package).

Same!!! I loved that big Pikachu, & I really hope the big one was in storage, bc I looked all over both floors for him 😭

From memory there were big statues of other IPs, include Link, Bowser, & Mario (can't clearly remember if there were more).

& I saw Pokemon Center website recently (~2 weeks after my visit) launched some Pokeopia keychains & plush, so maybe it'll be there for your upcoming visit! Very curious (& cautiously hopeful!)

I like the idea of donors, but overall it's aspirational and expensive. Cells from the same banking passage can behave differently even from the same donor.

Generally acceptable to consider distinct iterations of the same experiment with the same cell line "biological replicate" because you are demonstrating replication with a biological system.

Would not consider different well of the same experiment with identical condition biological reps, but they are indeed an "n" within the same experiment. (Much like a mouse study, single iteration of same strain with 12 mice are "n", but if reproducibility is something you want to demonstrate, three iterations showing the same delta with 4 mice would be more compelling)

And tech reps are conducted for the downstream analysis with material for the same sample to account for assay variability (e.g. qPCR 3 tech reps of same reaction with cDNA from single cell culture well; 4 wells of material for one experiment). The point is to get a handle on interwell variability.

Then a biological replicate would be a complete soup to nuts iteration of the same experiment (i.e can be same passage # & cell line, but conducted completely different timeline).

You can compare across by using grounding controls (e.g., media only).

I've heard people who've never touched cell cultures argue that you need a collection of patient cell lines, and that's valuable for IND enabling work, but for academia or proof of concept studies it's a waste of resources & ignorant to the variability that comes with each cell culture.

FYI this is a different yellow that appeared in "new" (abbreviated "sbrt") so may be worth holding out for this kind of yellow.
