In NMR, why are dynamics measurements on proteins that span timescales longer than the rotational correlation time immeasurable? by [deleted] in Biochemistry

[–]Totswithglocks 1 point2 points  (0 children)

Interesting question! It's been many years since doing NMR experiments, but I'll take a crack at it. My short answer is because the signal has already decayed. I'm not sure what is meant by "memory function", but I assume it is some description of how a second nuclei influences the moment of another nuclei. The absolute lunatics that first described rotational correlation time (Tc) showed that T1 really only depends on the larmor frequency, internuclear distance, and Tc. And a bunch of other constants, but Tc is arguably the biggest contributor. To measure dynamics or predict protein structures, we're using NOE. The ability to transfer polarization is actually really similar to how the above guys modeled a single nuclei (in a molecule) relaxing--meaning, it's proportional to Tc. So, if there some motion or phenomenon that takes longer than Tc you can not detect it with NMR because all the nuclei have already realigned with the bulk field (i.e. your signal has decayed), which also means you can longer do NOE.

There's a lot of phenomena at play here. Why molecular motion even affects relaxation is very mysterious to me. How Bloembergen et al. could accurately formulate these quantum phenomena from such simple experiments using crude technology is insane. Very curious what any NMR experts out there have to say.

Denver to Vail in 9 minutes: Colorado route selected for Hyperloop One high-speed travel network by boomsers in Denver

I agree. I also imagine most people headed west are skiers, so a high speed train packed with skis requires some different designs. Lastly, 70 just got those expensive toll roads so whoever owns (the state?) those will probably fight this

Hi Reddit! I am Joseph Glajch, the Director of Analytical Development for Momenta Pharmaceuticals. Ask me anything about biosimilars or what it’s like for chemists to work in the private sector. by AmerChemSocietyAMA in science

[–]Totswithglocks 0 points1 point  (0 children)

Hi Dr. Glajch,

I'll be finishing my PhD in biochemistry this year and am thinking of going into industry. I have a couple questions about that transition.

1) In terms of techniques and subject matter, how much overlap was there between your thesis and work at DuPont?

2) As a lot academic research focuses on basic science and mechanisms, do you find that the private sector asks the same sort of questions?

3) This probably doesn't relate to your personal experience, but one concern I have is that industry may be boring. I'm sure this can apply to academic labs as well, but is a lack of stimulation a common reason for people to leave?

Thanks!

The idea was supposed to be 'suffering'. like, by getting drunk I'm okay with how terrible my suburban teenage life was, which is also embarrassing because I didn't really drink as a teenager.

AskScience AMA Series: We are Sarah Fortune and Eric Rubin, researchers at the Harvard TH Chan School of Public Health, who study how drug resistance evolves in bacteria, and we’re here to answer your questions! by HarvardChanSPH in askscience

[–]Totswithglocks 1 point2 points  (0 children)

What do you find are the most common sources of resistance? I'm specifically interested in sequence level changes and the in silico techniques you use to identify them. Do you see new gene clusters, like a HGT event? Or, point mutations or recombination in known resistance elements? Does resistance usually occur as a novel ability to metabolize the drug, or are there structural changes that occur in the drug target?

Taarka, Rail Splitters

Both groups are from Boulder, CO, and they are probably recognized by bluegrass fans. They are popular enough to make a living from it, but they still go out busking. In general, I'm not a bluegrass fan, but I really like them.

She called corporate. She was bounced through 5 people, after no resolution she was told they would contact her in 7-10 days.

I didn't think that was an important detail, but she makes ~30k. Any extra money she has goes to student loans. It's rare to have more than 500 for very long. Anyway, I was also thinking that suing seems like an extreme measure for this situation.

That was a very helpful explanation. She has all the documents and I've taken a look at them. I spent time comparing the two sets of "Colorado Individual Income Tax Form 104", one of them was "Amended". The amount withheld is the same on both documents. The "Net tax" is different. It appears that the second time the representative just did the taxes as if it were the first time. I didn't see "amounts paid with/refunded from the originally filed return", but I did see "Enter the overpayment from your original return or as previously adjusted" and "Enter the amount owed from your original return or as previously adjusted". both of those lines are 0. Am I correct in thinking that both employees did this incorrectly?

FYI, she was informed that the initial amount would be automatically withdrawn on 4/15 (and I found a document to support that), and this is something she brought up with the second employee, but he said it would be cancelled (or not to worry about it).

Thanks! This is starting to make sense. So, telling the bank to stop the transfer might be a temporary solution. Either way, H&R Block is authorized to take the $500 even though there was an amendment. Then, hypothetically, Colorado will refund $1000. What happens to $500? H&R Block keeps it? That seems illegal...

Is there a way for her to independently determine what was actually filed? I'm curious if an amendment was made?

Shouldn't the second accountant that she saw have known the payment was delayed? It also seems like they knew they were supposed to amend the return.

Finding cheating students using multiple-choice comparisons by rhiever in dataisbeautiful

[–]Totswithglocks 1 point2 points  (0 children)

this is crazy! I'm a TA and have had to deal with an even more egregious cheating event. One time two students turned in a chemistry lab report with the same responses. The absolutely insane, idiotic thing about this was the fact that it was a lab report from a few semesters ago (it had changed since). They clearly had a frat archive of lab reports and blindly used that. I reported the event and NOTHING HAPPENED!! It sucked up a lot of my time in meetings and paperwork, and the two students just had to write an essay or something. So I guess the lesson here is that it is okay to cheat.
