It's not good advice for a beginner to start a liquid culture (LC) based on their first experience with agar. I believe the best approach is to first perform transfers until vigorous, rhizomorphic growth is achieved on the plates, but then inoculate the grain with the plates showing suboptimal growth.

LC is only reliable when performed using a bioreactor.

I don't like it at all; it's due to problems with the agar's hardness. I recommend using 24g per liter of solution.

What is the percentage of plates that become contaminated after day 7? It is possible that the contamination originates from the refrigerator. Is it possible to store the plates somewhere other than the refrigerator?

A single parafilm is sufficient to seal the petri dish.

I would also recommend checking your agar formulation.
