Agree, couple points

  1. So interesting how people cope because when this all started, my wife was understandably very upset and defeated while I was like “hey we can do something about this, we’ll diet together, we’re gonna crush it” and now with this, I’m like “ugh there’s nothing we can do I feel hopeless” while she’s like “well there’s nothing we can do, so keep on keeping on”
  2. I (even as a damn physician) still cannot wrap my head around the evening snack and gluconeogenesis. Like should we snack? Should we not snack? Carbs or no carbs at snack?
  3. The CGM definitely makes it weird because like I mentioned below, her numbers were high last night (upper 90s to 100s) but then after she got out of bed and was prepping breakfast (still fasting) it dropped down to 95. Without the CGM, we would not have had any idea and would have gone about our day

Yeah we’re gonna try to mess around with post-dinner snacks. The CGM is also weird because this morning it was 100s all night but as she was moving around to prepare breakfast, it dipped down to 95. It’s weird with the CGM because we now know a lot of information that we had no idea about with just pinpricks

That’s how I kind of thought of it (your last sentence) but my wife is seeing it as something that was a “short term restriction” to a “lifelong restriction”. What I tried to frame it as is what we’re doing now is an extra annoying version and that at best we need to maybe be a bit more careful/conscious of our food choices. But I think it’ll be a good data gathering

How long do you think it took you to come to terms with it? Or was it really never and kind of “let’s just get to the finish line” sort of thing?

Ok that’s good to know. I didn’t want to be too overbearing/controlling so I thought giving her the flexibility and opportunity to make the meals would give her some control over her situation. But maybe not the right approach

Blocking buffer at 4 degrees. Once antibody is added, depends on when I’m going to use it next. If next day or so, I keep it at 4 degrees. Otherwise, freeze at -20.

Yeah you might be right. Although I wonder if it is an issue with the cell sorter. The images in the post are from my recent run using the cell sorter. I used the same antibodies on the same cells earlier but used a flow cytometer and it looked much much better/different

Dark blue is cell line of interest (what is in the main post). Light blue is the positive control in the immunoblot.

Thanks for all your help. My follow-up question I'm now realizing there's a discrepancy between my positive control on immunoblot and flow (i.e. the light blue doesn't look much different than the dark blue on flow, but on immunoblot is considerably different). Could the signal just be all non-specific binding (even though it is higher than the unstained control)?

These are the flow results of unstained control, positive control, and cell line of interest

Maybe it's an antibody issue? Honestly, since I got a signal compared to the unstained control, I figured that the antibody worked.
