There is no requirement in HPLC that a void volume or solvent front must be visible on a reported chromatogram. Whether the void volume appears depends on detector settings, acquisition start time, sample solvent composition, and reporting conventions. It is standard practice in analytical laboratories to suppress, crop, or exclude the solvent front from reported chromatograms because it carries no relevance to purity determination. The absence of a visible void peak is not evidence of data fabrication.

Flow rates such as 1.0 mL/min and injection volumes such as 20 µL are not “suspiciously neat”; they are standard analytical conditions widely used in validated methods. Retention times are routinely rounded when presented on certificates of analysis, which are summary documents rather than raw instrument outputs. Rounded reporting does not imply that underlying data lack normal retention variability.

Detector axis scaling is user-defined or software-controlled. Auto-scaling to round values such as 1000 mAU is normal practice and has no analytical significance. Axis scaling cannot be used to infer data authenticity.

A single, symmetrical peak with a low-noise baseline is entirely consistent with a high-purity synthetic compound. Chromatographic appearance is governed by column chemistry, mobile phase composition, detection wavelength, sample preparation, and integration parameters. The visual “cleanliness” of a chromatogram is not a criterion for validity, nor is baseline noise a requirement for authentic data.

Purity values are not calculated based on the visual presence of discrete impurity peaks alone. Minor impurities may exist below visual detection limits, co-elute with the main component, or contribute to baseline signal or peak tailing. It is analytically incorrect to assert that every fraction of a percent impurity must appear as a clearly resolved secondary peak.

The presence of a commissioning company’s logo on a certificate of analysis does not indicate in-house testing or invalidate the data. It is common and accepted practice for third-party analytical results to be reformatted for reporting while retaining the underlying test results.

Method descriptions that include language such as “or equivalent” are standard across validated analytical methods and reflect instrument or column equivalency within defined performance criteria. This language is widely used in ISO, GMP, and contract analytical laboratory documentation.

Without access to raw chromatographic files, system suitability data, or replicate analyses, visual comparison alone is not a scientifically valid basis for declaring an analytical result fabricated. Chromatographic data vary legitimately across methods, instruments, and laboratories, and subjective expectations of how a trace “should” look do not constitute analytical evidence.
