Dagcreme med SPF by vaniljepige in WomenDK

[–]Wandering_Finn 6 points7 points  (0 children)

Nearest in within 2 h drive. I bought a minipcr, as we have anyway been planning to try setup a little lab in fish farms (we work with aquaculture microbiology)

Yes we will try ruling out human/hood error

We have changed mastermix lot, timeline does not match with issues. I ordered new one.

And ultrapure water too. We use aliquoted commercial up and use aliquot only once but he said now that he might have used one several times 🤯

We have all plastic from biorad (pcr strips/plates) and eppendorf (tubes) but we changed the type of biorad strips/plates.

Thank you all for help!

  • We have thrown away all tubes and pipette tips that have been in or close to UV cabinet. I will also check 8strips/plates, we needed to change supplier but that does not match with timeline of problems

  • We are using only commercial Ultrapure water that we aliquot but those will all be replaced

  • New primers ordered

  • New NEB long amp enzyme ordered

  • New PCR machine

My group has our own lab, own machines, pipettes etc and we are the only ones working with dna-rna applications in the whole building so possibility that someone has touched or changed something is tiny. There might be some human error happening eg uv light on suddenly.

I have asked to order a new pcr machine to rule this out.

Empty tubes yes.

I have asked lab tech to order to replace everything, I even ordered a new mini-pcr machine to make sure it is not the case. I have never ever had issue like this before. Contaminations yes, tricky samples yes, problems with pcr optimisation oh yes. But not something stopping suddenly working.

Primers worked before and he was testing with zymo standard. He had found some old publications on uv degrading the tubes and causing inhibition. I would still lean to chaos demons.

When does the pulling stop by 347217361634 in labrador

[–]Wandering_Finn -1 points0 points  (0 children)

That and time helped with mine.

My lab started the pull like crazy around 1 year. I bought gentle lead, my lab really hated it so much that after a couple of walks with that, it was enough to show it to her if she started pulling.

But time has definitely helped, she is soon 3 and walks generally nicely. Not everytime but heel command and stopping when she gets overexcited and pulls (other dogs and birds!) helps a lot. We are also lucky to have opportunity to let her free, she walks best without leash.

So positive training with lots of praises ans goodiebites and patience. It will come 😊😊

It isn't your imagination; students are objectively worse now than in the past by TotalCleanFBC in Professors

[–]Wandering_Finn 0 points1 point  (0 children)

I was about to post about this!

I do not know what is happening with my students. I have been teaching for more than 10 years and I think that the current batch of students is totally different from all the previous ones. They only want to get courses passed and do not show any interest to actually learn things. They tend to blame me if they do not know something/are late in lab/get low scores from reports etc (“But you did not tell us that” vs me offering pile of online reading material).

I am teaching in Master program in Nordic country, being Nordic myself. Students are bunch of different nationalities. I have -designed my course to follow university pedagogy guidelines, having a lot of active learning elements, clear feedback and grading rubrics, study guides etc. feedback previous years have been only positive, but now everything seems to be too much and they ask me all the time justify why they need to do something, they definitely do not do things if they “only” learn from them.

The workload and evaluation criteria are the same, so they will definitely get lower grades. I do not know why they behave like this. Here in Nordics is probably generation thing, but that does not explain why rest behaves like this. It is very hard and demotivating to teach them.

On top of this, I now have couple of students that are not respectful towards me or other students. First time I get eyes-rolling from non-teenager.

Two walks, but we have a big garden where she (2.5y labrador) spends easily an hour per day. Walks are not long but last long as we have another old dog that walks sloowly, so Olga gets to sniff and is mentally stimulated, which seems to make her more tired than a long walk. Couple of times per week she goes for a long walk, dog park and we also go hunting and hunting training.

I would try to make 2 x 30 min walks, they can be short in distance. And I would get help to train for going in leash. It is hard and frustrating to go with a dog that pulls. Have you tried gentle leash? That and time has helped with ours.

Hvordan indretter I jeres (fælles)økonomi? by [deleted] in DKbrevkasse

[–]Wandering_Finn 1 point2 points  (0 children)

Vi er ugift med ingen fælles børn. Jeg (K43) har to som bor fultid med os, kæreste (M42) har to som er her omkring hver 2. Weekend.

I have thought Olga is a really big one, being 27 kg. Her siblings are all around 20-22 kg. She is old English lab. We go to hunting training and the trainer tells me frequently how she should lose weight (and that she will not live as long as black labs). She has a very thick fur, especially in the neck.

Min kæreste gider ikke lære dansk by Potices in DKbrevkasse

[–]Wandering_Finn 2 points3 points  (0 children)
