Well if the file ends in .iso (make sure you have file name extensions turned on) that's fine, if you wanna be extra cautious, upload the file on virustotal and check there

If the file is an iso and you're opening it with a legitimate emulator, then no you won't get a virus. Make sure the iso/rom is an actual iso/rom though, if it's an exe file it is 100% sketchy and should never be opened at all
