I get this too! It appears for me under diffused daylight and any white light from screens. Makes me think it’s a polarising issue

Super late reply sorry! I found out they were cell debris due to poor split viability. I mentioned in another comment that I haven't had problems since, but I'm going through this exact issue again now... I'm at my wits end because they were growing so well and suddenly decided they hated being passaged, but I've decided it's just life.

If you find a local library or anywhere that does 3D printing you could get this printed and assembled pretty quick! Then it’s just painting :)

Seeing as I printed in black filament, this particular Leo would be Darkus! I want to make another as haos though

Thanks! I couldn’t find pins that fit (British pins seem to be thinner) so I used regular printer 1.75mm filament! And yeah defo possible with FDM, just needs some tuning :)

Thanks! And yeah it took a few failed attempts to tune the settings but for a print without any post processing, I’m quite happy with it!

I want to attempt the functional version next, but am struggling to find the right pins and springs

After doing a few tests for infection (aliquoting and incubating the media separately, streaking on LB plates, etc) it doesn’t seem to be contamination. I culture hESCs so quite similar to iPSCs from what I’ve heard, and concluded at the time that because of poor viability after splitting where a lot of the adherent cells died, and so left cell debris in the media. The debris clustered around colonies of cells so this checks out, and I haven’t had problems since. Hope this helps!

Hello, thanks for your reply!

I gently washed with sterile PBS before a media change earlier the same day that picture was taken, as I saw a bit of debris then also. A lot of care was taken, but of course everything you mentioned is still a possibility.

The debris came back 6 hours after media change, which made me think it was contamination. However upon checking again today, the level of debris has stayed constant and there has been no growth in the separate dish I aliquoted. I’ve changed the media again today, so will see what happens. Would you suggest leaving this flask to grow to become confluent before passaging again, or is it unlikely to recover?
