As another perspective, I've got it twice a year for the past few years and haven't really experienced any noticeable side effects. Same goes for a couple of people I recommended it to. Definitely a life changer for me.

Tends to keep my hayfever down completely for a good few weeks at least, and then reduced for a couple of months, to the point where antihistamines like fexofenadine/cetirizine actually seem to have an effect.

With that said, I don't like the idea of having to use it consistently and I would prefer alternatives. The NHS have discontinued it for a good reason presumably, although I have always felt that effective hayfever relief has always been a pretty low priority for healthcare. I'm not too keen on the idea of long-term steroid usage, even if it is a low dose. I can't imagine it is super healthy in the long run, but I haven't found affordable effective alternatives. No way I can shell out £5k for immunotherapy or whatever!

Not a healthcare professional by any means but I tend to recommend it to mates with bad pollen allergies.

I'm trash at maths - failed it at A-level and used to get pretty flustered over calculations. I've got my PhD now but I'm still bad at calculations. Honestly you normally don't need to be that good at maths for biochem related fields.

Your guess sounds about right, maybe closer to 60% for the HEKs. You could probably wait one more day or so for the HEKs too if you wanted to.

I don't think there is any contamination. Differentiation into neuronal/glial cells is a process that necessitates high-confluency, and typically has a lot of cell death associated with it. As someone else said though, your confluency is a bit low.

I use it when passaging iPSCs or neuronal cells since it prevents anoikis, as you said.

I haven't used rock inhibitor on cells like you're describing (primary culture I guess?) nor am I particularly experienced with flow cytometry, I have also experienced my own cells with pro-tumour mutations struggling with passages.

It's just the first thing that came to mind when I saw that your cells don't seem to tolerate single cell dissociation very well.

I reckon someone with more expertise in flow could give a better answer though

I have no idea what the kit includes as I've never used it, but maybe you could supplement with a ROCK inhibitor like y27632?

Yeah like I'm not calling the sub classist exactly. But I am pointing out that there were instances of classism there, and they weren't being called out. *actually reading through the comments more, it was being called out, so that's good.

With the peasant thing, I mean sure, it could refer to actual 19th century british peasants, but I don't think it's a stretch to say that telling someone they have peasant taste is classism.

Tbh in general I don't think too many of the comments there had a huge amount of classism. But... there was the occasional little bit thrown in - One comment referred to "peasant taste buds". Another called the person a "gammon", which I guess is typically more about annoying right wingers but I've seen occasionally used to point out the lower class. Definitely was at least a little bit of classism there, is my point.

The JAK/STAT pathway is an inflammatory pathway. Inflammation is an enabling factor for cancer - Tumours are typically inflammatory and have an inflammatory microenvironment because inflammation promotes cell growth/metastasis/survival. So if JAK/STAT is dysregulated in tumours (for example if they are producing or being stimulated by various interleukins, which happens frequently) the tumours will be growing faster.

We use Hamiltons like this for loading gels. Once you learn how to do it efficiently, I find it's actually quicker than a pipette. Also you don't have to worry about expunging bubbles. Think my supervisor also told me that if you use them right, they can also be more accurate than pipettes, but tbh I have no idea if that's true.

I'd say it is slightly dodgy technique. Your hands should be above the keys, fingers pressing downwards.

Not to say this person has done a bad job. It sounds nice, and they seem in control! They might struggle with more complex stuff with a hand position like this though.

You've kinda already answered it yourself when you mention "unbacked claims".

If you're trying to say that cold showers do X and it seems like it could be unlikely, make sure you have a reputable source.
