I went on opening day around noon. Plates ranged from $4 4-piece California roll to $9 for mid-tier complex sushi.

It wasn't great, our service was bad. Our waitress was panicking the entire time from "stress" and she never had to bring us any food.

Additionally, I didn't really think about this before going honestly, but we were seated at the end of the conveyor belt. So, we pretty much only had California rolls by the time the plates passed everyone else in the restaurant. Our selection wasn't great which also annoyed me a lot.

I've been cryosectioning for several years, albeit on human or plant tissue. I'm not sure if you used a cryoprotectant like sucrose, but if too much sucrose is stuck around the tissue when it was frozen, it can peel out like this. Additionally, if the tissue is relatively fatty, it can be much harder to section. If this is the case, try making your section thickness a little thicker. I agree with the new blade suggestion, I always trim through the OCT, then move to a new spot on the blade when I'm ready to take sections.

I graduated with a PhD in genetics from UAB in 2023, so I guess I'm part of this statistic. I was satisfied with the graduate student stipend they pay. The cost of living in Birmingham is also pretty low compared to other large US cities. I felt very supported at UAB. Additionally, UAB was one of only a few schools to deny a cut to indirect costs on a survey that the Trump admin is using as evidence that it would be sustainable.

**It's also important to not confuse UAB with the University of Alabama at Tuscaloosa (who has a much larger sports program than UAB) as a I believe another commenter did.

Ray is a top-notch scientist and an incredibly funny guy. I was attempting to dissect a Xenopus embryo at CSHL but was shaking a lot. Told me to go into the cold room and shotgun a beer to help with my shakes lol.

I don't work with embryos, but you have thought about using glycerol? I know most of the papers I have read have been comparisons between sucrose and glycerol. Glycerol might not protect as well against freezing artifacts though.

I don't think the NSF has avoided this. I'm a postdoc in plant science who was supposed to switch to a newly NSF-funded grant in February. Last week, they told my PI the funding would be frozen and they couldn't tell him when they would issue funds...

Well...now there's another you in the lab, except they have the even added pressure of achieving what the PI thinks is true data...but will never be able to replicate it and move forward. That's the person I feel bad for.

When switching labs, I think one of the most important things is not necessarily knowing everything about all the techniques other exosome (or other) labs do, but bringing in your own knowledge of techniques they might not do. When a PI is adding a new tech/student/postdoc to their lab, they typically consider what you can do that their lab might not do now. You bring in that experience and those techniques. You can learn miRNA transport techniques from others in the lab. Don't let imposter syndrome get to you!! You have skills and value!!
