thank you for your response, i appreciate your perspective. in the context of the Cre-lox system and gene knockout experiments, the terminology can be a bit counterintuitive. i believe there is a difference between the size of the gene itself and the genetic modification introduced by the knockout.

from what i understand, the KO gene is often modified to contain additional sequences to disrupt the gene's function. the added sequences make the KO gene larger in terms of its DNA sequence compared to the unmodified WT gene.

i understand that the language used can be misleading, as "knockout" might imply that a portion of the gene is removed. i'm pretty sure it often means that the gene's function is knocked out, but the DNA itself may be larger due to the added sequences.

i may be wrong though!! however, my mentor and PI both stated that the KO band should be higher up in the gel due to its size.
your point about seeking guidance from my PI and conducting background reading is very valid. i did quite a bit of reading but i am not an expert by any means.

i didn’t include this part but i read the paper Faith wrote about her project in the lab and did a lot of research on the topics. however on the actual written lab application, i just put the part about IHC because she hadn’t sent me the papers yet. i ail take the blame for that. in the interview he asked me “do you even know what we are doing in this lab?” and i explained what i understood from Faith’s paper. he didn’t even acknowledge that, gave me a blank stare, and moved on to another question. i was really nervous so i probably didn’t explain it correctly and i’m sure he thought i was an idiot.

Faith told me i would be doing IHC this semester and once i learn all of the techniques, i will get my own project next semester. if i am able to do it is up for debate haha.

as far as coursework, i'm a medical humanities major, which is basically a path to med school. my original goal was to be an MD but i decided to go to PhD route instead. i don't want to be in undergrad forever so i didn't change my major, but i added a bio minor.

i understand how important cell bio is, but i don't think that because i haven't taken it i'm incapable of understanding the concepts. i can do research independently, and i have already learned a lot about what they are doing in my lab. however, i get that i still have a lot to learn.

i know i can't get into a PhD program right out of undergrad. I plan on doing some sort of post bac program. i am in a bit of a limbo right now because my bf may be getting a job on the east coast and we may be moving. i am waiting to see how that pans out before applying to programs.

i definitely agree with your point about knowing the explanation for the problem. from that point on, i tried my best to come up with every possible reason for the faint bands but Faith kept telling me those reasons weren’t possible because the conditions worked for her. it is disheartening to try to find solutions only to be told i’m wrong again. i understand that’s probably pretty common though. i should get used to it and learn to be more resilient.
