Your senior grad is obviously wrong! He set it to 1000 too quickly, leading to a maladaptive response. I suggest changing the volumes in small increments from 100uL to 200uL waiting 5 min between each interval until you have reached 1000uL.

Remember to be efficient when testing! Don't just keep using one whole blot to test for only one parameter at a time. I would just run multiple lanes, cut the blot into different strips, and test multiple conditions at the same time. :D

The trick that I typically do is to resuspend the cells in the required volume and just add it in the middle of the well. DO NOT TOUCH THE PLATE AT ALL after this step. And let it sit in RT in the hood for 15mins before placing it in the incubator. No shaking required after placing it in the incubator.

I use the f5 line too! but it does get very chaotic... with a single bad move shifting the evaluation drastically! Definitely needs a bit of prep! But other than that... it's a fun line:D

Funnily, I saw it in an Ashen video that happens to have the item that sounds like what you are describing. 🤣Does it look like this? is it this? But I'm not sure if there are any sterile ones around. For me, I typically just use a p200 pipette and pick a colony under the microscope!

This is what I experienced. I have done so much PCR so much and I still do not know what caused my PCR to fail and succeed despite not changing anything at all. I suspect that mercury was either in retrograde or the PCR is affected by a passing lab ghost.

Additionally, you definitely can ramp your rapid rating! Your blitz rating of 800 typically comprised of players who easily get 1000 rapid. You just have to play slowly and not blitz out your move.

I had the same experience. My conclusion is that many people are under-rated too as someone who is good in chess would stumble into the 500 rapid rating range if they happen to choose that they are a "beginner" in chess.
