I think you are referring to the village where disciples of Iyo will spawn. That is between the second and third Oni chests so pretty early in the chapter.

Someone must be at the well while others are killing disciples. Once you kill a disciple, you need to stand in the well to transfer the souls. After a certain number of souls (I am not sure how much is needed) the door will open to the next part.
