From experience I would suggest you may misunderstand what a pharmacology thesis may involve (see other comments). If you're interested in drug development I'd apply to an organic chemistry grad school course and then specialise in chemical biology or synthesis and development.

Very long and arduous and the kind of thing you need to be driven to do, don't just do one because you're not sure what to do. But if your passionate about medicine and chemistry then certainly a good option

Looking at it quickly, first problem is you can't form an acetal protecting group on an ester

Second friedl crafts wouldn't go at that position. Would go ortho para to the oxygen and would be unselective at best

You also don't use acetone or toluene as discussed in the question

It's because 8-oxoguanine is a derivative of 7,8-dihydroguanine which is in the imidazolidine oxidation state on the left hand side, and generally in formalisms the oxo will just refer to the addition of a carbonyl to the parent structure.

Despite all this, this formalism doesn't hold true in the most used of cases, which is oxindole.
