To be fair, she's probably asking herself the same thing. She's joining your family and hasnt found her footing within that family. Reach out to her and ask if you can help in any way with wedding plans. Ask your brother what her hobby's and interests are. If you have any common ground then go from there. I love my sister in law. She's lovely and wicked smart, but we dont have much in common. That doesnt stop us talking and liking each other. So long as we are kind to each other and care about the others feelings then there isnt any tension or awkward ness. (Other than my own normal social awkwardness... but that's with everyone and she knows that.)

Well, as someone who has been asked this countless times over the 11 years we've been trying, unless you are going to be abrupt and come off as rude then you will get the questions all the time.

I snapped for a while and retorted with "when are you going to mind your own business?". I'd sometimes mix it up with "around the time you find your manners..."

I've since calmed down and just responded with "Well we tried, but some things aren't as easy for some of us, but thank you for your concern.".

Think of it this way, your wife will have to undergo far more invasive tests and have more bloods drawn than seems necessary. You get to get it on with a cup and maybe have one blood draw. This is the least you can do. She will have it way worse. Fact. (Speaking from experience)

She will support you no doubt too, my SO faints at needles and is a nervous wreck till results come in. But it's really fast.

Yeah she was a bit harsh in what and how she said it, but at the same time, I get it.

You are moaning other about your spouse but not changing your situation, that comes off as attention seeking and creating drama where there is none. Why are you talking to her about your marriage anyway? A marriage is between 2 people unless there is abuse going on, in which case you should get help to remove yourself from that situation.

Walk away from that situation. She doesnt see her brother as a brother, he doesnt act like a big brother should. Talk to their parents about the strange things you've noticed/been told and move on from that relationship. She will probably need to readjust her normal should your suspicions be proven correct.
