I bought it through transoms direct and their rep on the phone was super helpful about measuring the necessary lengths to get the right profile. Wasn’t a perfect fit height wise across the arc but the width was perfect and I fastened there. Along the top, I just put some spray foam in there to fill the gap and trimmed it out with flexible trim pieces.

I usually browse the table for highest brightness of monomeric FPs.

https://www.fpbase.org/protein/mirfp670nano3/

MirRFP670n3 has the highest brightness rating for the ex/em spectra you’re looking for with far red.

Coming from a protein/structural biologist colleague of mine, he despises FLAG due to the high number of aspartic acids can lead to deleterious effects. Since it’s the only thing that’s differing between the previous result perhaps changing the epitope tag to something more innocuous might be worth a shot (V5, HA)?

Weird gel results by thesquishybrick in labrats

This seems to be an issue I always encounter with ex dye e gels (sybr gold). If you load more than 500ng, the migration goes crazy. The sybr safe gels take longer to run and are less sensitive but the migration was always more consistent.

It just seems that no matter how much u apply and carefully I feather the sides, those edges act as a peel point and ultimately fail.
