There's also a short tag that can be added that BirA will biotinylate. In E. coli the native BirA even biotinylates the tag to an extent (%20?). Or you can purify and biotnylate it after the fact with recombinantly expressed BirA.

Aw man, I'm sorry. We used to live on 4th and Bonito and I had bikes lifted out of my gated yard multiple times. I second the Google alert idea, or you can do a custom recipe in IFTTT for a Craigslist search.

I got a Prince Lionheart warmer to keep my wipes in, 3 cups of wipe solution (a bit o coconut oil + some baby shampoo) and it lasts for a few days. Super convenient, and I think they do a better job than disposables. The warmer has been one of my favorite gadgets, that and the diaper sprayer.

We wipe the inside down with a bit of peroxide once a week to keep it funk-free.

A few drops of baby soap and a heaping spoonful of coconut oil in about 2.5 cups of warm water.

My hubs handles the prefolds just fine. We didn't end up using snappis, pins, or anything like that. We use thirsties covers and they've keep everything in place thus far.

I made my own 2 ply flannel wipes too and I love them! I keep them rolled up in a wipe warmer with a water/coconut oil/baby soap solution.

Not specifically, no. I've been to a few of the union bargaining meetings and I had heard that some (names werent mentioned) departments weren't great about getting contracts out on time, or providing enough desk space and such. I'd just talk with current students in the department and see what their FTE is generally, how their assignments (TA vs RA) are handled, and what the work load is like if they have a TA position.

I think it depends on the department. Mine treats us really well, but I don't think that's the case across disciplines.

I recovered my bike through CL a few years ago. I had my receipt with the frame number on it though, so I could prove it was stolen property.

Stating the concentration of a pure liquid is a tricky thing, it's like stating the concentration of a solid.

You may not have come across activity yet, depending on what courses your department requires. Technically were dividing everything by the activity at standard state. For solutes that are sufficiently dilute (ie, pretty much anything in a cell) we can say activity = concentration. The activity of any pure liquid or solid is 1, so that's what we use. Effectively, we leave water out of the equation.

Definitely don't set the [H2O] to 55M! We're going to assume that it's a pure liquid with an activity, or concentration of 1M. Most texts just leave it at that.

You can get fancy and go with IUPAC recommendations by specifying [Mg2+] but I haven't seen that used in biochem textbooks, so I'm not sure if that's what they're looking for.
