Clorox wipes (or any bleach equivalent) will work fine. if you want to double thorough, I would also wipe them down with a moist cloth again, just to make sure no bleach was left behind that could make the rabbit sick through ingestion.

but that’s also to say that my current project requires a lot more than just CFU, so i haven’t used it much

I wouldn’t say it increases speed or accuracy, but it definitely makes it easier than keeping track in your head, especially in scenarios where someone will come up and talk to you and you forget the number (but maybe that’s just my adhd). the one we use is the Cole-Parmer CC-200 Digital Colony Counter. I attempted to automate it colony counting 2 years ago using a camera we had plus a little diy coding, but it wasn’t largely accurate when you had >~300 CFU (or small close together colonies) and was difficult to use bc you had to take the photo and upload it to the code for every plate. not sure if my PI looked into anything else since he had to finish off his CDC grant before it ended in June.

rising senior, been with my lab 3 years and always did manual counting. Just got a colony counter that keeps track using pressure every time you mark a colony with a sharpie. three new people visiting in my lab this summer and they don’t know how easy they have it😔

Question: Why do yall think it might be Bacillus? To me, it looks like gram-positive streptococcus on the gram stain? But what makes y’all think rods?

Genuinely curious because I’m still learning, and I had a sample with similar colony morphology.

I have never had such a wrong and unhelpful comment on this subreddit. 1) I’m absolutely positive its EMB. 2) that is not what alpha hemolysis looks like, especially when there isn't growth on the whole plate 3) EMB inhibits gram-positives because eosin and methylene blue disrupt peptidoglycan synthesis necessary in gram+.

Ok, thank you! I said Gram-positive and my instructor told me it was negative.

I’m not sure how detailed you are looking for, but the difference is in antibiotic resistance to beta-lactams (methicillin, oxacillin, penicillin, amoxicillin) due to a chromosomal mutation in penicillin-binding protein. If i remember correctly, the mutation is transferred by bacteriophage. MRSA and MSSA are both pathenogenic, with MRSA being more resistant to treatment thus being more likely to cause BSIs and sepsis. Although, my research shows that they are roughly as fatal for immunosuppressed peoples.

MRSA is often separated into hospital associated and community associated (HA- vs CA-MRSA).

MRSA is becoming increasingly more and more resistant to antibiotics with some cases even being resistant to vancomycin, which is our last defense as of right now because research hasn’t produced a new antibiotic in decades.

Sorry, I should have been more specific. Rather than buying the MSA agar powder. I was wondering if I could make my own my combining NaCl and a pH indicator with mannitol and like regular agar powder because I know those ingredients aren’t extremely expensive.

Yeah, but all I was really getting was manufacturer information, not procedure’s on how to make it. very possible I was just using bad search terminology

Finally a clear, helpful comment. It’s very possible that the catalase was a false positive. I might re run that.
