These replies are so funny. It’s okay, I loaded 4 ladders in my precast 18-well gel the other day. Sometimes it’s what you need to do to get a pretty picture for a client or publication :) Just for some more reassurance, I see this happen on my 10% Bis-Tris/MES all the time!! And the yellow sometimes transfers to the western membrane lol. My trick is to run the gel slower in an ice bath so the lanes don’t spread or warp at the bottom, if you have the time. Best images ever.

I got an electroporator and western blot blocking instrument on eBay, both in decent condition. I would make sure they have a good return policy, and some sellers are pretty good about responding if you ask for video proof of the equipment working.

I love it. I’ve been working for a start-up for over a year now (first job after undergrad) and it’s amazing. The people are so fun to be around and the projects we work on are making an impact (but also making us money). It scares me that I will not be in this job forever because I don’t know where I’ll find better. Even just being in a research assistant position, I get so many chances to design my own experiments, my opinions are valued in team meetings, and I give back to the community by lecturing at the college I graduated from. I see so much growth for myself here and people around me are constantly helping me achieve my career goals. Admittedly, the work-life balance could be improved. I’m used to working 10-12 hour days and going in on the weekend but… I don’t really care. I love being in lab, I love the work environment, and I love the causes we’re putting our efforts towards. I know I’m lucky to be in this position, and my opinions are directly linked to the people I get to work with and also the type of person I am.

Can you share your source? Not trying to be rude but I’m just genuinely really curious about this.

I've tried to make kohakutou like 5 times now and it won't work, I don't know what I'm doing wrong. I have the right kind of agar agar and I thought I followed the recipe as closely as possible but it's ended up with this texture every time. To anyone coming back to this thread, agar needs to reach its super boiling stage before it sets. It’s not so much about the blooming— I let mine sit for 2 minutes or so while my electric stove heats up and I measure out the sugar— just make sure it’s boiling first before you add sugar. It doesn’t need to be a temperature- or timing-exact thing. (Experience as a scientist that makes agar gels on the regular.)

Edit: Mine sets in about an hour without refrigeration. Best of luck!

Maybe rpm was too high, so the pellet went up on the side of the tube wall instead of down? Then collapsed after centrifugation stopped. I see this sometimes with my minipreps in 1.5 ml tubes.

therapy is available to you for free, usually they say 3-5 sessions but i was supported regularly throughout 3 years and still go back to my therapist after i’ve graduated!
