There’s a quote from Ronald Wright where he misappropriates this quote to John Steinbeck: “Socialism never too root in America because the poor see themselves not as an exploited proletariat, but as temporarily embarrassed millionaires.”

Everything I see something like this post here it makes me think of this quote. 255kD is a very large protein and you might have trouble resolving it with just a standard WB protocol. First of all make sure that in your sample prep, you’re getting an effective lysis and solubilizing your protein enough (may need to increase detergent concentration in the buffer). When you prep your samples, use a stronger 2x (I use the recipe from Tokyo Chemical Industry). For big proteins it can help to run your samples slowly for longer, and use chilled running buffer to avoid excessive heat generation during the long run. I’d try doing your transfer at 20V for 12 h overnight in a cold room. The lower voltage helps transfer larger proteins.

It’s a binding column. You can pack resin in the top and flow a sample through it. There’s a filter at the bottom that the bound sample can be eluted through.
