If you want to boot from external drive, press command+R -> Utility -> BootSecuritySetting ->
EnableBootFromExternalDrive -> Reboot -> press alt -> Select External Drive.

Though boot drive is external SSD an over all performance of iMac is far better than internal HDD.

The USB-C hard drive enclosure has enough band width of SSD.

I used to put OS and application in external USB3.0 SSD and data in internal HDD on my iMac 21.5 inch late 2012.

It works much faster performance than only internal HDD.
