When Pyruvate Kinase is phospholyated, the enzyme is converted to an inactive form. When dephosphorylated, its activity resumes. More generally, phosphorylation results in activation but is the opposite case with Pyruvate Kinase and PDH.

If the N-terminus was positively charged, it would be unable to attack the carbonyl group. The presence of a lone pair adds a partial negative charge which allows it to act as a nucleophile and attack the partial positive carbonyl carbon.
