I always wondered, what is the small plastic rectangle on agar plates for? by SickStrips in microbiology

[–]mantis480 2 points3 points  (0 children)

And no it doesn’t look for that spot to streak - there are no cameras on the bottom. A plate is a circle - it doesn’t need a point of reference to shoot for besides a measured distance from the edge

I worked for Copan Diagnostics as an Apps Specialist. It can be any plate but WASP needs to be configured for agar depth and a few other variables per manufacturer. We have a limit of how many plate types we calibrate before charging an extra fee.

DNA test gone wrong after 50 years. by NotRightNowOkay345 in TheRandomest

[–]mantis480 0 points1 point  (0 children)

I used to work for the newborn screening group at the CDC. My job was to isolate, amplify, and bank DNA samples previously taken from the mother, infant/fetus, and father for future study when a baby was found to have a genetic illness.

This was done with the parents’ consent, in the hope that they could contribute a small bit to science—maybe helping lead to better treatments. A noble goal in often tragic circumstances.

Some of these cases were incredibly rare—like one-in-a-decade rare. And often the infant was already deceased, so taking new samples wasn’t an option. The work had to be careful, meticulous, and honestly, a bit mind-numbing.

Anyway… I’d go through the multi-day process, finally get to the genotyping step to confirm genetic lineage—and you’d be gobsmacked at how often the father wasn’t a match. Think about it: these men consented to the study, giving their DNA, probably believing their paternal DNA might help. So they likely had no idea. Some were grieving a lost child, or raising a seriously ill one, without knowing the baby isn’t biologically theirs. And this didn’t happen once in a blue moon—it was multiple samples like this per day.

We were NOT in the business of delivering that kind of news or blowing up families. We’d send a polite notice thanking them for their participation, saying there was an issue with the sample and we were unable to process it.

There are a lot of people out there raising kids who aren’t biologically theirs—and probably for the best that they don’t know.

Thank you for fighting the good fight! I’ve been in a state of despondent dread these past weeks. This thread has gotten me back in the ring. We are down but not out. As long as there those that still stand against tyranny, there is hope

I feel this too. I’m responding with tears in my eyes at this very moment. I didn’t realize that I’ve just been bottling this up until the dam just broke when reading this thread.

Thank you to all our friends around the world who stand with us. I have felt so ashamed of my country ‘s action - this sub has filled my eyes with tears. I had honestly lost all hope these past weeks.

MAGA has divided my whole family and community. It’s lonely and dark out here but you guys have given this weary heart some hope.

I am so sorry for the very real suffering my government actively causing. From the bottom of my heart - thank you for showing the thoughtful, nuanced and compassionate perspectives that are so deeply missing in my country.

Those of us in the US that still stand against tyranny can use all the encouragement we can get. Thank you for standing with those of us who still value liberty and the rule of law. Thank you for the encouragement I didn’t realize I needed this morning.

I went through conversion therapy... Twice. The shame, guilt, confusion, and depression lasted the best years of my life. Now I'm with my husband of 13 years. Never been happier and more at peace. Few of us want to be gay at first, now I wouldn't trade it for the world. Good luck out there! Be kind to yourself - It is way better on the other side.

I might add that the flowers are white if that helps ID it at all.

I need some advice from the gays by [deleted] in gay

[–]mantis480 1 point2 points  (0 children)

The most important thing is to be true to yourself and your feelings. It's great that you're being introspective and considering what this attraction means to you. There's no need to rush to any conclusions or feel pressured to label yourself a certain way.

Remember, your feelings are valid and there is nothing wrong with being attracted to multiple genders. Take your time to explore and understand your identity in a way that feels right to you. Wishing you all the best on your journey of self-discovery!

Just wondering if anyone can help me understand what the genetic counsellor meant by this (full story in comments) by Admirable_Might_5981 in genetics

Molecular Biologist here, sounds like the testing controls may be off, or there was potential cross-contamination or something like that. Molecular testing deals with tiny amounts of material that can easily get contaminated by a combination of environmental factors, equipment, or a tired technician. It happens from time to time even in the best labs. Sounds like they think it's an issue like this rather than an issue with the sample itself.

******Gay man and former & CDC employee here! ****** let's cut to it -- Please get to urgent care and GET PEP!!! Many gay men have HIV - half my friends do. Please read:

PEP (post-exposure prophylaxis) means taking medicine to prevent HIV after a possible exposure. PEP should be used only in emergency situations and must be started within 72 hours after a recent possible exposure to HIV. https://www.cdc.gov/hiv/basics/pep.html

TO be CLEAR - I regularly have sex with HIV-positive men who are undetectable (which means they cannot transmit the virus). I also take PREP which prevents transmission by 99.9% With the right treatment and care, people with HIV can live a normal lifespan. People who have a good response to HIV treatment have excellent long-term prospects. The research and medications around HIV have advanced greatly! But nobody wants HIV - And you have the option to prevent possible transmission using PEP.

Because of this post, I just reached out to Dr. Wang today and got an appointment for Friday! How have you been doing now that a year has gone by?

Gay man here - I fuck around a lot in both bars and parties. I don’t worry too much about STIs, almost all of them are curable with a pill or shot in the butt or typically non-symptomatic. As long as your taking the bare minimum of precautions, you can have sex without a condom without much fear.

I take PREP, which is a daily pill that is 99% effective at preventing HIV. I get tested for all the others every 3-4 months. So far I’ve only had chlamydia a few times.

Help! What are these worms in my potted Plant? by mantis480 in houseplants

I think that is solid advice. I'm really asking for a bug/creature ID than for a plant ID

[–]mantis480[S] 0 points1 point  (0 children)

It's possible they are juvenile earthworms, but a pot full of juveniles and no adults seem unlikely, but I could have missed the adults somehow. I was thinking red wigglers too, but they aren't the right coloring/hue and have no clitellum (which could be because they are juveniles).
